Abstract

The method includes: well mixing collected corn silage to use as a sample; accurately weighing 20g of the silage sample, and adding into a 500ml glass beaker; adding 180ml of distilled water, stirring for 1min, standing at the room temperature for 30min and filtering; determining the ammoniacal nitrogen content of extract liquid. The sample pretreatment method has advantages that high accuracy in determination isachieved, and a standard deviation of three replicate samples can be controlled within 0.23%; distilled water serving as extraction solution can be more easily obtained as compared with deionized water and ultrapure water; the extraction temperature is the room temperature, and the temperature environment requirement in an extraction period can be lowered; an extraction concentration is determined, namely the 20g of the silage sample is extracted in the 180ml of distilled water; the extraction time is 30min and much shorter than existing common extraction time which is 24h.
