Problems solved by technology

The tissue culture rapid propagation system is limited by the explants of the female parent. The contamination rate of the explants is too high after simple disinfection, and browning is easy to occur after excessive disinfection. It is difficult to establish a sterile line

Embodiment Construction

[0019] method embodiment

[0020] 1. Selection and disinfection of explants: select the inflorescence rachis that has grown for about 20 days as the explants, wash them clean, and then disinfect them. The disinfection process is: rinse them with clean water, rinse them once with Amway detergent, and shake them on a shaker for 15 minutes. -20min; after rinsing with running water for 20-30min, disinfect with 75% ethanol for 10-20s on the aseptic operating table, immediately pour 0.1% mercuric chloride for disinfection for 12min, and finally wash 4 times with sterile water, shaking 3- After 5 minutes, the sterilized inflorescence rachis was cut into 2 cm long segments, and a total of 100 inflorescence rachis segments were inserted.

[0021] 2. Induction of callus: take the explant fragments obtained in step (1), and inoculate them in the induction medium (invert the slant of the medium in advance) on a slant to induce callus. The culture temperature is 25±2°C, first cultured in ...

Abstract

The invention discloses a tissue culture propagation method for Heuchera micrantha variety 'Obsidian'. The method comprises following steps: (1), an explant is selected and disinfected; (2), after treated rachis is cut, callus induction and culture are performed; (3), callus is subjected to differentiation for induction of adventitious buds; (4), the adventitious buds are subjected to subculture multiplication to form seedlings; (5), test-tube plantlets obtained after subculture multiplication are subjected to rooting culture; (6), seedling hardening culture is performed. The Heuchera micrantha variety 'Obsidian' is subjected to tissue culture with the propagation method, the adventitious bud proliferation rate in one subculture period (35d) is 10 times or higher, the propagation speed ishigh, and the problem about seedling breeding is effectively solved.

Description

technical field [0001] The present invention specifically relates to a kind of alum root ( Heuchera micrantha ) Propagation method in tissue culture. Background technique [0002] alum root ( Heuchera micrantha ), also known as Coral Bell, is a perennial herbaceous flower of the saxifrage family. It has a wide range of horticultural varieties and colorful leaves, and the color of the leaves will change in different seasons, environments and temperatures. It is an ideal perennial flower border material. At present, alum root plants are deeply loved by home gardening small flower lovers, and the e-commerce sales of some varieties are in short supply. In addition, in gardens, it is also used for understory flower borders, ground covers, garden greening, etc., and is an excellent perennial color-leaf flower in a shade environment. [0003] Alum root plants like partial shade, are tolerant to full light, low temperature, and avoid excessive humidity. They are suitable for pla...
