Embodiment

[0028] experimental method

[0029] (1) Tissue samples

[0030] 100 tissue samples from patients with colorectal cancer were collected. The isolated tissue samples were immediately quick-frozen with liquid nitrogen and stored in an ultra-low temperature refrigerator at -80°C. The tissue sample was confirmed to be colorectal cancer tissue by histopathological examination.

[0031] (2) Cell culture

[0032] SW620, LS174T and normal human colonic epithelial cells FHC were from Shanghai Cell Bank of Chinese Academy of Sciences. CRC cells were cultured in RPMI 1640 medium containing 10% fetal bovine serum and cultured at a constant temperature. The constant temperature culture conditions are 5% CO2, 37°C.

[0033] (3) RNA extraction and fluorescence real-time quantitative PCR

[0034] Take 500 mg of tissue sample, grind it into powder with liquid nitrogen and transfer it to EP tube, add 1 mL of LTrizol, shake and mix, and let stand on ice for 5 min. Centrifuge at 12 000 rpm at 4°C for 10 ...

Abstract

The invention relates to the technical field of molecular biology, in particular to application of microRNA and a target gene thereof in colorectal cancer. According to studies, the expression level of miR-149 in colorectal cancer patients and CRC cells is significantly lower than that of normal tissues and colorectal epithelial cells. The low expression of the miR-149 suggests poor prognosis in the colorectal cancer patients. The miR-149 promotes the metastasis of the colorectal cancer and the migration of the colorectal cancer cells by acting on STAT3. The effect of miR-149 and the target gene of the miR-149 on the CRC cells is analyzed by the in vitro cell method. Results show that miR-149 inhibits the proliferation, invasion and migration of the colorectal cancer cells by targeting STAT3. The miR-149 can also promote the apoptosis of the colorectal cancer cells by targeting STAT3, and can be used as a target for the treatment of the colorectal cancer.

Description

Technical field [0001] The invention relates to the technical field of molecular biology, in particular to the application of microRNA and its target gene in colorectal cancer. Background technique [0002] Colorectal cancer (CRC) is the third-ranked cancer in the world and is a malignant tumor derived from glandular epithelium. Due to the continuous improvement of the country’s economic and material levels, people’s eating habits and diet structure have undergone major changes, and the aging population continues to grow, resulting in a significant increase in the incidence of colorectal cancer in my country, which greatly harms the health of residents and affects Its quality of life. [0003] MicroRNAs (miRNAs) belong to a class of non-coding RNAs that are found in eukaryotic cells and consist of 19-25 nucleotides. miR-149 is a miRNA that has been studied in recent years. As a tumor suppressor gene, it participates in the regulation of the occurrence and development of liver canc...
