Abstract

The invention discloses a method of further screening salt tolerant tomato from seedlings of tissue culture of certain salinity under hydroponics condition by utilizing tissue culturing in combination with nutrient solution culturing to screen the salt tolerant tomato. The method consists of the following steps: (1) sample seeds provided are sterilized and inoculated in sterilized water and germinated; (2) germinated seeds that are acquired from step (1) are respectively inoculated on MS culture medium for NaCl stress treatment; (3) salt damage investigation and calculation of salt stress indexes; (4) seedlings of tissue culture under NaCl concentration of 90mmol.L<-1> are transported into a greenhouse and transplanted into the nutrient solution of the concentration of 90mmol.L<-1> after seedling recovering. Seedlings are harvested 10 days later, and all indexes of the seedlings are recorded. The method of the invention can assess salt resistance of tomatoes of various breeds more accurately, and is of great significance for providing theoretical basis for screening and utilizing of salt resistant tomato seed resource and establishing salt resistance culture.
