Problems solved by technology

However, there are many varieties of onions, and the proliferation effects of different varieties of onions on different components of the culture medium are very different, which directly affects the proliferation effect; moreover, vitrification and aging of tissue materials often occur during the cultivation process, which greatly limits Training effect

Embodiment 1

[0050] Example 1. A method for onion tissue culture, the steps are as follows:

[0051] (1) Store the harvested fresh onion bulbs in a 4°C environment for 3 weeks;

[0052] (2) Peel off the outer skin of the refrigerated onion balls until the diameter is 2cm, then disinfect it, and rinse it with sterile water after disinfection;

[0053] (3) Under aseptic conditions, cut the sterilized onion bulbs to explants with a bulb disc thickness of 1 mm, a bulb length and width of 4 mm, and a height of 0.5 mm, which are close to the bulb disc. Make a "well"-shaped scratch on the upper surface, and then inoculate the explants into onion proliferation medium I or II;

[0054] (4) After 20-30 days of cultivation, obtain aseptic materials with several lateral buds; Cut off or break off the lateral buds from the bulb and transfer them to onion proliferation medium III or IV for 20-30 days to obtain aseptic materials that have grown clustered buds, and then they can be multiplied every 20-...

[0090] Example 2. A method for onion tissue culture, the steps are as follows:

[0091] (1) Store the harvested fresh onion balls in a 5°C environment for 4 weeks;

[0092] (2) Peel the refrigerated onion balls until the diameter is 2-3cm, then take the onion balls under aseptic conditions, shake and soak them in alcohol with more than 75% alcohol for 30-60 seconds, and then use 0.1% by mass of liters Shake and soak the mercury solution for 10-15 minutes, disinfect it, and rinse it with sterile water after disinfection;

[0093] (3) Under aseptic conditions, cut the sterilized onion bulbs to explants with a bulb disc thickness of 2 mm, a bulb length and width of 5 mm and a height of 1 mm close to the bulb disc, and then use a blade on the bulb. Make "well"-shaped scratches on the surface, and then inoculate the explants into onion proliferation medium I or II;

Abstract

The invention relates to a method for culturing onion tissues, which is characterized by comprising the following steps of: refrigerating harvested fresh onion bulbs, peeling onionskins until the diameter of each onion bulb is between 2 and 3 centimeters, and sterilizing the peeled onion bulbs; excising the sterilized onion bulbs under the aseptic condition to form explants, and inoculating the explants to an onion proliferation culture medium I or II; culturing to obtain aseptic materials on which a plurality of lateral buds grow; performing propagation once every 20 to 30 days; and transferring the propagated strong seedlings of an onion individual plant to a rooting culture medium for culturing to obtain live onion seedlings. In the method, onion varieties are subjected to tissue culture by different culture media, wherein the proliferation rate of the onion varieties is improved by adding Hyponex; and in the proliferation process, the proliferation rate can be improved by 3 to 5 percent by the culture medium added with paclobutrazol appropriately, and the phenomena of the vitrification of tissue culture seedlings and the aging of tissue materials can be reduced or relieved to a certain extent by the culture medium added with phloroglucinol appropriately.

Description

technical field [0001] The invention relates to a method for cultivating plants, in particular to a method for culturing onion tissue. Background technique [0002] The onion industry is an industry with significant benefits. At present, onions have become one of the most important foreign exchange-earning vegetables in my country. Many farmers have achieved poverty alleviation and prosperity through onions. In order to meet domestic and foreign market demands, the demand for fine varieties of onions is increasing day by day. Since onions have only been introduced into my country for a hundred years, and there are few onion varieties in our country, it is necessary to import a large number of seeds just for the introduction of varieties and the selection of conventional varieties, and the cost is very high; in order to speed up the breeding of hybrid varieties, it is necessary to obtain the sterile line of onions and maintain the system. Whether it is the preservation of e...
