Abstract

The invention relates to the technical field of culture preservation of edible fungus cultivation and especially relates to a boletus aereus stock culture low-temperature preservation method which includes following steps: (1) cultivating mycelia of boletus aereus fruiting body on a solid culture medium to obtain a mycelia block; (2) soaking the mycelia block in sterilized distilled water in a sterilization container, sealing the container, and allowing the mycelia block to stand for 10-30 days at 10-15 DEG C; and (3) performing low-temperature preservation at 3.5-4.5 DEG C. According to the method, the stock culture is preserved in a step-by-step refrigerating manner. A test proves that the stock culture preserved through the method is equal to the first generation stock culture in mycelia activity and bacterial strain cultivation characters. The method can keep the stock culture activity and excellent characters of the boletus aereus, can solve the problem of middle- and short-period simple bacterial strain preservation of the stock culture, and can provide a powerful guarantee of large-scale cultivation.
