Abstract

The invention discloses a culture method for in-vitro expansion of hematopoietic stem cells and homing ability thereof. The culture method includes steps: (1), adopting a culture medium A to culture the hematopoietic stem cells for 10-20 days, wherein the culture medium A is obtained by adding growth factors like TPO, Il6, SCF and Flt3 into a basal culture medium; (2), after the above culturing procedures, separating out 50% cultured cells, and adopting a culture medium B to culture the cultured cells for 1-12 days to obtain HPC culture liquid, wherein the rest cultured cells are HSC culture liquid, and the culture medium B is obtained by adding growth factors TPO, IL6, SCF and Flt3 into the basal culture medium; (3), mixing the HPC culture liquid with the HSC culture liquid, culturing, and centrifuging to harvest cells. The growth factors are combined with micromolecular compounds to culture and expand the hematopoietic stem cells, so that number of hematopoietic progenitor cells is increased, glycosylation is adopted to improve homing ability of the hematopoietic stem cells, and transgenic technology is not applied, so that the culture method is high in safety.
