The invention belongs to the technical field of plant tissue culture, and discloses a Paeonia fulva tissue culture and rapid propagation method for inducing cluster buds by using immature embryos, which comprises the following steps: collecting fresh seeds at a specific development stage, sterilizing, and stripping embryos aseptically; inoculating the embryos into a germination culture medium containing 6-BA, IAA and GA3 to start culture; after radicle tail ends of the germinating embryos are cut off, the embryos are directionally inoculated to a subculture medium according to the original polarity, and sterile seedlings are obtained; transferring the seedlings into an induction culture medium containing 6-BA, IAA and charcoal to induce cluster buds, and further proliferating; cutting robust tender shoots, removing cotyledons, and inoculating into a rooting culture medium containing IBA (indolebutyric acid), NAA (naphthylacetic acid) and charcoal to induce rooting; and finally, carrying out strong seedling culture and domestication transplanting to obtain a survived plant. According to the method, the deep dormancy obstacle of the seeds is broken through, efficient and stable regeneration from embryos to seedlings is achieved, the breeding cycle is short, the rooting and transplanting survival rate is high, and the method is suitable for large-scale protection and production of rare resources.

A method to alleviate cadmium toxicity during buckwheat germination and seedling stages

ActiveCN117397418Bpromote germinationImprove germination rateBiotechnologyPlanting seed
This invention relates to a method for promoting the germination of buckwheat seeds under cadmium stress and alleviating cadmium toxicity during the seedling stage. The method includes treating buckwheat seeds with a melatonin reagent before germination. The method involves contacting the plant seeds with a melatonin solution at a concentration of 100-500 μmol / L. The method provided by this invention can effectively improve the germination potential and germination rate of buckwheat seeds under cadmium stress, enabling uniform and rapid germination. This invention solves the important problem of how to ensure normal germination of buckwheat in cadmium-contaminated soil in agricultural production and has significant application value in production. This is likely because pretreatment with a melatonin reagent can induce the expression of antioxidant enzyme activity, scavenge the large number of free radicals generated by cadmium stress, alleviate oxidative stress, reduce oxidative damage, and maintain a low level of membrane lipid peroxidation in the seeds, thereby promoting normal seed germination.
