Embodiment 1

[0056] The in vitro cultivation of embodiment 1, Rhodiola rosea clump

[0057] 1. Selection and disinfection of explants

[0058] Rhodioladumulosa (Franch.) S.H.Fu] was collected from Dongling Mountain in Beijing at an altitude of about 2000m in July 2012, and was confirmed according to the morphological characteristics of Rhodioladumulosa (Franch.) in "Flora of China", and the real object was consistent with the name . Choose the young and tender stem section of Rhodiola clump, rinse it with tap water for 20mins, use 75% (volume fraction) alcohol surface disinfection for 10s on the ultra-clean workbench, rinse it with sterile water for 3 times, and then wash it with 0.1% (mass fraction) ) mercury liter disinfection for 2mins, rinse with sterile water 3 to 5 times.

[0059] 2. Induction culture of callus

[0060] Cut the sterile stem segments that have been sterilized in step 1 into small segments with a length of 0.5 cm, and inoculate them into callus induction medium (MS+...

Abstract

The invention discloses an in-vitro culturing method for rhodiola dumulosa. The in-vitro culturing method for the rhodiola dumulosa comprises the following steps that 1, stem segments of the rhodiola dumulosa are inoculated onto a callus tissue inducing medium to be cultured to obtain callus tissue; 2, the callus tissue obtained in the step 1 is inoculated onto an adventitious bud inducing medium to be cultured to obtain adventitious buds; 3, the adventitious buds obtained in the step 2 are inoculated onto an adventitious bud multiplication medium to be cultured to obtain multiple buds; 4, the multiple buds obtained in the step 3 are inoculated onto a rooting medium to be cultured to obtain regeneration seedlings. In-vitro culturing of the rhodiola dumulosa is performed by adopting the method, the rooting rate can reach 100 percent, the average rooting number is about 10-15, and the root length is 2-3 cm; regeneration plants are transplanted after rooting is achieved, and the survival rate reaches 90 percent. According to the in-vitro culturing method for the rhodiola dumulosa, an in-vitro culturing regeneration system of the rhodiola dumulosa is built, and the important significance is achieved for effectively protecting and utilizing wild rhodiola dumulosa resources.

Description

technical field [0001] The invention belongs to the field of plant tissue culture, and relates to an in vitro culture method of Rhodiola rosea clumps. Background technique [0002] Rhodioladumulosa (Franch.) S.H.Fu, also known as Fengweiqi, is a perennial herb of Crassulaceae. Produced in northwestern Sichuan, Qinghai, Gansu, Shaanxi, Hubei, Shanxi, Hebei, Inner Mongolia, Jilin, on hillside rocks at an altitude of 1600-3900 meters. [0003] Xiaocong Rhodiola is a precious Tibetan medicinal material, also known as "Saul Marbu". Modern medicine has confirmed that Xiaocong Rhodiola has the function of "adapting to the original state" similar to the traditional Chinese medicine "strengthening the body", that is, promoting the morbidity caused by different causes to move toward the original state. Special regulation functions for normal state transitions. Its nourishing, strengthening, disease-preventing and anti-aging effects are rare among known tonics. Rhodiola chrysalis is...
