Embodiment 1

[0044] Example 1. A method for identifying low-fat broiler chickens using the expression level of TCF21 gene mRNA.

[0045] (1) Primer design and synthesis

[0046] According to the chicken TCF21 mRNA sequence (Genebank accession number: NM_001277711), primers were designed to amplify the CDS region of the TCF21 gene. The upstream primer was TCF21-F: 5'-ACGCTGCCAACGCAAGGG-3', and the downstream primer was TCF21-R: 5'-TGTTCACCACTTCTTTCAGGTCACTC-3' ;According to the chicken TBP mRNA sequence (Genebank accession number: NM_205103): design primers to amplify the CDS region of TBP, the upstream primer is TBP-F: 5'-GCGTTTTGCTGCTGTTATTATGAG-3', and the downstream primer is TBP-R: 5'-TCCTTGCTGCCAGTCTGGAC- 3', the primers were synthesized by Yingwei Jieji (Shanghai) Trading Co., Ltd.

Abstract

Belonging to the field of animal molecular genetics, the invention provides an identification method for low-fat broiler chickens by TCF21 gene mRNA expression. Directed at the problem of low-fat broiler chicken identification, the invention provides the identification method for low-fat broiler chickens by TCF21 gene mRNA expression. The method includes the steps of: designing and synthesizing aprimer pair for amplifying the CDS region of TCF21 gene and a primer pair for amplifying the CDS region of TBP gene; extracting the total RNA of abdominal adipose tissues of a to-be-detected broiler chicken and conducting reverse transcription to obtain cDNA; conducting fluorescent quantitative PCR on the abdominal adipose tissue cDNA of the to-be-detected broiler chicken; and when the relative expression of TCF21 gene mRNA in the abdominal adipose tissues of the to-be-detected broiler chicken is smaller than 0.125, identifying the to-be-detected broiler chicken as low-fat broiler chicken, with the relative expression being calculated with TBP gene as the internal reference. The method provided by the invention can be used for low-fat broiler chicken identification.

Description

technical field [0001] The invention belongs to the field of animal molecular genetics, in particular to a method for identifying low-fat broilers by using gene expression. Background technique [0002] After long-term selective breeding, the growth rate and meat yield of broilers have been significantly improved. However, broiler breeding is facing new challenges: with rapid growth, broiler physiological discomforts and related diseases have increased significantly, such as excessive body fat accumulation, ascites syndrome, sudden death syndrome, leg diseases, decreased immune function, etc. These problems cause huge economic losses to broiler producers. Excessive accumulation of body fat (especially abdominal fat) in broilers has become a prominent problem. Excessive fat deposition in broilers has many disadvantages: (1) significantly reduces feed conversion efficiency, because fat deposition per unit weight consumes three times more energy than lean meat deposition per ...
