Embodiment 1

[0040] Example 1: Preparation of active components from the stem of Akebia trifoliata

[0041] Dried Akebia ( Akebia trifoliata(Thunb.) Koidz.) stem 100kg, sliced, placed in a 2T capacity extraction tank, added 10 times the weight of 75% ethanol solution, refluxed for 3 times, filtered, concentrated under reduced pressure to dryness to obtain the total extract. The total extract was dispersed by adding water 1:2, extracted with equal volumes of dichloromethane and n-butanol successively, each extracted three times, and the n-butanol extract was collected. For the n-butanol extract, wash with an equal volume of ammonia test solution and water successively, each washing twice, discard the washing solution, collect the n-butanol extract, concentrate under reduced pressure to dryness to obtain the n-butanol extract. The n-butanol liquid extract is carried out to silica gel column chromatography, 1.5 times of silica gel is mixed and sampled and put on the column, and the chlorofo...

Embodiment 2

[0042] Example 2: Preparation of Active Components from the Root of Akebia clover

[0043] Dried Akebia ( Akebia trifoliata (Thunb.) Koidz.) root 100kg, pulverized, placed in a 2T capacity extraction tank, added 12 times the weight of 95% methanol solution, refluxed twice, filtered, concentrated under reduced pressure to dryness to obtain the total extract. The total extract was dispersed with water at 1:4, extracted with equal volumes of dichloromethane and n-butanol in turn, each extracted four times, and the n-butanol extract was collected. For the n-butanol extract, wash with an equal volume of ammonia test solution and water successively, each washing three times, discard the washing solution, collect the n-butanol extract, concentrate under reduced pressure to dryness to obtain the n-butanol extract. The n-butanol liquid extract is carried out to silica gel column chromatography, 1.5 times of silica gel is mixed and sampled and put on the column, and the dichlorometha...

Embodiment 3

[0044] Example 3: Preparation of active components from the leaves of Akebia trifoliata

[0045] Dried Akebia ( Akebia trifoliata (Thunb.) Koidz.) Leaves 100kg, placed in a 2T capacity extraction tank, added 5 times the weight of 90% ethanol solution, refluxed 5 times, filtered, concentrated under reduced pressure to dryness to obtain the total extract. The total extract was dispersed with water at 1:5, extracted with equal volumes of dichloromethane and n-butanol successively, each extracted twice, and the n-butanol extract was collected. For the n-butanol extract, wash with an equal volume of ammonia test solution and water in turn, each washing four times, discard the washing solution, collect the n-butanol extract, concentrate under reduced pressure to dryness to obtain the n-butanol extract. The n-butanol liquid extract is carried out to silica gel column chromatography, 1.5 times of silica gel is mixed and sampled and put on the column, successively use the chloroform...

Abstract

The invention relates to the field of natural medicines, and particularly discloses an active component with a diuretic effect extracted from akebia trifoliata, a preparation method and an applicationof the component. The active component is extracted from akebia trifoliata, and the method includes the following steps: (1) preparing an extract; (2) extracting a solvent; (3) washing and concentrating the extract; and (4) purifying by silica gel column chromatography. The active component obtained by the above method steps contains 40-90% of total saponin by mass; the total saponin comprises four saponins of Saponin PH, Akemisaponins E, Saponin PJ1 and Scheffoleoside A, and the four saponins account for more than 60% of the total saponin in the active component by mass; and the active component extracted from akebia trifoliata contains the four saponins which have mutual synergistic effects, do not need component separation and purification, and are directly used for preparing diureticdrugs.

Description

technical field [0001] The invention belongs to the field of natural medicines, and in particular relates to an active component with diuretic effect extracted from Akebia trifoliata, a preparation method and an application. Background technique [0002] Clover Akebia ( Akebia trifoliata (Thunb.) Koidz . ), commonly known as August melon vine, three-leaf vine, and August hawthorn, belongs to the family Akebia, which belongs to deciduous woody vines. Its dried canes have the effects of inducing diuresis and treating stranguria, clearing heart-fire and relieving restlessness, dredging menstrual flow and releasing breast milk, and are used for stranguria syndrome, edema, upset urine, sores on the mouth and tongue, amenorrhea with less milk, and damp-heat arthralgia. Since ancient times, Akebia trifoliate has been used to treat gonorrhea. It is recorded in "Medicine Properties": "It is mainly used to treat five stranguria, facilitate urination, and open and close the grid. It ...
