Abstract

The invention discloses a short peptide for promoting nuclear translocation of TFEB, a linear short peptide based on the short peptide and its application for alleviating cerebral ischemic injury, and belongs to the technical field of biomedicine. The short peptide disclosed in the present invention is a short peptide fused with the functional region of TFEB protein and 14-3-3 protein, and its nucleotide sequence is shown in SEQ.ID.NO.1. The invention also discloses a linear short peptide protecting the above short peptide, which is formed by fusion of a membrane-penetrating peptide and the above short peptide. The present invention verifies through experiments that the synthesized fusion peptide can regulate TFEB nuclear translocation, promote CMA and reduce cell damage. Therefore, its application as a new therapeutic target for alleviating neuron death and improving cerebral ischemic injury is disclosed.

Description

technical field [0001] The invention belongs to the technical field of biomedicine, and relates to a short peptide that promotes TFEB nuclear translocation, enhances chaperone-mediated autophagy (CMA) and relieves nerve damage and its application, and specifically relates to a method for promoting TFEB nuclear translocation Short peptides and linear short peptides based on them and their application in alleviating cerebral ischemic injury. Background technique [0002] Stroke is a serious threat to human life and health. There are approximately 16.9 million new stroke patients worldwide each year, and 5.9 million patients die of stroke-related diseases; another 33 million stroke patients need long-term treatment and care, which brings heavy burdens to society and families. Burden (Lancet, 2017). tPA (Tissue plasminogen activator) is currently the only drug approved by the US FDA for the treatment of ischemic stroke, but its treatment time window is short and increases the r...
