Embodiment Construction

[0024] The present invention selects 10 adolescent male cynomolgus monkeys (21-52 months old) and divides them into two groups according to age and body weight. Subjects in the Chronic Unpredictable Stimuli group (CUMS, n=5) were exposed to several unpredictable stimuli for 5 cycles. The control group (CON, n=5) was treated according to daily routine throughout the experiment. Behavior, body weight, and sucrose preference of all monkeys were measured at baseline and at the end of each stimulation period. In addition, the apple craving test (AAT), human trespasser test (HIT), and cortisol levels in hair and blood were measured at the end of the study.

[0025] So the concrete steps of the present invention are as follows:

[0026] 10 adolescent male cynomolgus monkeys (21-52 months old) were selected and divided into chronic unpredictable mild stimulation group (denoted as CUMS group, n=5) and control group (denoted as CON group, n=5) according to age and body weight. =5), s...

Abstract

The invention discloses a construction method of a depression model for crab-eating macaques at adolescence and juvenility. The construction method comprises the steps of selecting 10 male crab-eatingmacaques at the adolescence, and dividing the selected crab-eating macaques into a chronic unpredictability stimulus group and a control group; performing 7-day daily treatment, and after adaptationis completed, performing 8-day baseline test; performing 5-cycle experiment while performing daily treatment on the chronic unpredictability stimulus group, including 7-day chronic unpredictable lightstimulus and 4-day behavior observation, performing daily treatment on the control group in the whole experiment process, and after every cycle is completed, also performing 4-day behavior observation; after the whole experiment cycle is completed, also performing apple desiring experiment and human intruder experiment on the chronic unpredictability stimulus group and the control group to respectively test the depression action and the anxiety-like action of the crab-eating macaques; collecting hair and plasma and performing cortisol content test; and finally, performing contrast with adolescent depression patients by a plasma metabonomics method. The model can provide reference for researching a potential mechanism of depression of teenagers in the future.

Description

technical field [0001] The invention relates to the field of animal models, in particular to a method for constructing a depression model of young cynomolgus monkeys. Background technique [0002] Adolescent depression specifically refers to depression that occurs during adolescence, and is a group of common mental illnesses characterized by significant and persistent depression, loss of interest, social withdrawal, and decline in ability. The latest epidemiological survey has pointed out that the prevalence rate of depression in adolescents (13-18 years old) is about 4-5%. Adolescent major depressive disorder is a common and serious mental illness with a high risk of suicide and other disorders. Global statistics show that suicides due to depression peak in people aged 10-24. [0003] The contribution of nonhuman primates to stress provides an excellent model for studying the mechanisms of depression. Existing studies on the pathogenesis of depression are often based on ...
