Problems solved by technology

[0010] However, the aerobic denitrifying bacteria found so far are mainly isolated from soil, domestic sewage and industrial sewage, and few are directly isolated from marine aquaculture biofilters.
In addition, it has been found that bacteria with aerobic denitrification function isolated from seawater have low nitrogen removal efficiency. For example, the aerobic denitrification bacterium Kocuriapolaris AD7 isolated by Wang Xiaojing et al. has a nitrate removal rate of only 80.70%. With the accumulation of a large amount of nitrite, the actual denitrification efficiency is very low, which restricts the development of denitrification treatment process for seawater pollution.

Embodiment 2

[0064] The identification of marine aerobic denitrifying halomonas described in embodiment 2

[0065] 1. Physiological and biochemical experiments of strains

[0066] The strains were cultured in BTB medium from the seed tube for physiological and biochemical identification. The physiological and biochemical characteristics of the strains were detected by Mérieux reagent strips, and the operation was strictly followed the instructions of API 20NE reagent strips and API ZYM reagent strips, and the experimental results were observed and recorded.

[0067] In the physiological and biochemical experiments, the Gram staining of the bacterial strain was negative, and it was a Gram-negative bacterium. The test results with the API 20NE reagent strip are shown in Table 2-1. It can be seen that the bacterial strain can reduce nitrate to nitrite, And reduce nitrite to ammonia or nitrogen. The strain indole test was negative, and it was negative in urease and arginine hydrolase tests. ...

Embodiment 3

[0083] Aerobic denitrification performance of halomonas described in embodiment 3 under different carbon source conditions

[0084] Add 100mL of denitrification performance test medium to 250mL Erlenmeyer flasks respectively, the culture solution uses glucose, trisodium citrate, sodium succinate and sodium acetate as the sole carbon source respectively, and keeps the carbon-nitrogen ratio at 15, to the culture medium Inoculate 3mL of the strain activation culture solution (the following inoculum volume is 3mL) into the culture medium, and then incubate at 30°C and 100r / min with constant temperature and shaking. Three parallels were set up for each treatment group. NO in the medium was detected every 12h 3 - -N, NO 2 - -N and TN concentration changes, experimental results such as figure 1 , figure 2 with image 3 shown.

[0085] Depend on figure 1 with figure 2 It can be seen that under the condition that the carbon source is sodium acetate at 72h, NO 3 - - The lo...

Abstract

Marine high-efficiency aerobic denitrifying halomonas and its application and method for treating aquaculture wastewater, the preservation name of the strain: Halomonash hydrothermalis Z8 , depository unit: China Center for Type Culture Collection, deposit date: March 25, 2019, deposit number: CCTCC NO: M 2019197. The strain has efficient denitrification ability under aerobic conditions. Through the determination of aerobic denitrification performance, the nitrate removal rate of the strain was 99.05% and the total nitrogen removal rate was 84.17% after 72 hours of culture. The strain was expanded and cultured and inoculated into the bioreactor to simulate the denitrification effect of the biofilter. The results showed that the film was quickly and efficiently formed, the denitrification efficiency was high, and the nitrite accumulation was not obvious. The denitrification performance was good. The invention provides a new theoretical basis for constructing a new denitrification mechanism in a seawater circulating water system, thereby further improving the biological filtration function of the circulating water system.

Description

technical field [0001] The invention relates to the technical field of microorganisms, in particular to a marine aerobic denitrifying halomonas, its application and a method for treating aquaculture wastewater. Background technique [0002] Seawater recirculating aquaculture is a high-density and intensive aquaculture model. The key to the system is the treatment of aquaculture wastewater to achieve the purpose of safe recycling, and biological treatment is the core of the recirculating water treatment system. [0003] The existing biological filtration is to convert the highly toxic ammonia nitrogen and nitrite nitrogen in the aquaculture water into less toxic nitrate. Due to the high dissolved oxygen concentration in the circulating aquaculture water, the traditional anaerobic denitrification cannot be carried out. This leads to the accumulation of nitrate nitrogen in the aquaculture water, which will bring potential harm to farmed fish after accumulation to a certain exte...
