Embodiment 1

[0023] A method for screening multiple sclerosis disease biomarkers, comprising the steps of:

[0024] (1) Obtain the dataset GSE21942 from the GEO database (http: / / www.ncbi.nlm.nih.gov / geo / ), the platform is GPL570, and the sample contains peripheral blood samples from 15 normal people and 14 MS patients For nuclear cell gene expression profiles, the samples were standardized using MAS5;

[0025] (2) x and y represent two genes respectively, using the formula (|(x(i)+y(i)) / (x(j)+y(j))-1|<δ(i,j=1 ,2,...,N), remove the x or y genes with unchanged expression, screen out the mutually inhibited gene pairs (x, y), and obtain multiple sclerosis disease biomarkers; where: δ represents gene x The degree of approximation to y is taken as δ=0.08 in the experiment; x(i) represents the gene expression of the i-th sample of gene x, and y(i) represents the gene expression of the i-th sample of gene y (assuming a total of N samples); the screening is implemented by writing codes in MATLAB....

Abstract

The invention belongs to the technical field of bioinformatics and particularly discloses a method of screening multiple sclerosis biomarkers. The method includes: acquiring a data set GSE21942 from an GEO database, and subjecting a sample to MAS5 standardization by a GPL570 platform; removing x or y gene expressions invariable according to a formula: (|(x(i)+y(i)) / (x(j)+y(j))-1|(delta(i, j=1, 2,..., N), and screening out mutually inhibiting gene pairs (x, y) to obtain a multiple sclerosis biomarker. Delta refers to degree of approximation between genes x and y, and is 0.08 in experiments; x(i) refers to gene expression quantity of the i-th sample of the gene x; y(i) refers to gene expression quantity of the i-th sample of the gene y. The mutually inhibiting gene pairs in mononuclear cells in peripheral blood of both normal people and MS patients to obtain the multiple sclerosis biomarker; by applying the screened multiple sclerosis biomarker in the study and analysis of biological significance, real conditions of the patients can be indicated.

Description

【Technical field】 [0001] The invention belongs to the technical field of bioinformatics, in particular to a method for screening multiple sclerosis biomarkers. 【Background technique】 [0002] Multiple sclerosis (MS) is an autoimmune disease characterized by inflammatory demyelinating lesions of the white matter of the central nervous system, the pathogenesis of which has not yet been fully elucidated. The clinical manifestations of the disease are multiple lesions scattered in the white matter of the central nervous system and remission and recurrence during the course of the disease. The brain, brainstem, cerebellum, and spinal cord of the patient may be involved simultaneously or sequentially. The clinical manifestations are various: such as limb weakness; Acute optic neuritis or retrobulbar optic neuritis, vision loss; ataxia; mental symptoms are mostly depression, irritability and bad temper. [0003] Currently, biomarkers, cerebrospinal fluid examination, magnetic reso...
