Embodiment 1

[0098] experimental method

[0099] The production method of human liver buds is based on "Methods for producing tissues and internal organs" (WO2013 / 047639).

[0100] (1) Production of human liver endoderm cells

[0101] Activin, Wnt 3a, and NaB were added to a serum-free medium to culture human iPS cells (as an example, human skin-derived TkDA3hiPSC clone (provided by the University of Tokyo)) to obtain endoderm-lineage cells (definitive endoderm: DE).

[0102] (2) Production of human liver buds

[0103] The resulting endoderm cells, vascular endothelial cells (from human umbilical vein endothelial cells) (Lonza, Basel, Switzerland) and undifferentiated mesenchymal cells (from human bone marrow mesenchymal stem cells) (Lonza , Basel, Switzerland) were mixed at a ratio of 10:7:1 (400:280:40 per organ bud) to produce human liver buds (size 120-150 μm).

[0104] (3) HBV infection to human liver buds

[0105] The human liver buds produced in (2) were cultured in a medium c...

Abstract

Provided is a virus infection model with which the shortcomings of conventional hepatitis virus infection models are overcome. The present invention provides a method for building a virus infection model with which it is possible to reproduce a viral life cycle, including infecting, with the virus, a cell aggregate formed by culturing tissue or organ cells in vitro. The virus infection model is avirus infection model with which it is possible to reproduce a viral life cycle including a cell aggregate infected with the virus, wherein the cell aggregate is formed by culturing tissue or organ cells in vitro. The present invention provides a method for screening for substances possessing antiviral activity by using the virus infection model.

Description

technical field [0001] The present invention relates to a virus infection model, its production method and its utilization. Background technique [0002] In recent years, attention has been paid to the method of creating human functional cells useful for the creation of new drug screening and regenerative medicine through induction of differentiation using pluripotent stem cells such as iPS cells that have the ability to differentiate into various functional cells. So far, the research group of the inventors of the present invention has established a method for reconstructing human tissues / organs with an orderly three-dimensional structure composed of vascular endothelial cells and mesenchymal cells that can be observed in adult tissues (not Patent document 1: Takebe T.et.al., Nature (2013) VOL.499, p.481-485; Patent document 1: Method for making human tissue / internal organs WO2013 / 047639). [0003] On the other hand, the infection model of the hepatitis virus that people s...
