Problems solved by technology

[0003] From a technical point of view, there are currently technical bottlenecks in the classification and detection of chemical health hazards, such as low throughput, long time, cumbersome operation, and lack of multi-component detection equipment. In the detection of endocrine disrupting effects, it is urgent to improve the detection throughput and shorten the detection time. Improve the automation of routine testing and solve the problem of metabolic activation in vitro
Therefore, the current detection of chemical endocrine disrupting activity has technical problems such as low throughput, long test cycle, few toxic targets in one test, and low connotation of toxic effect spectrum.

Embodiment 1

[0044] 1. Prokaryotic expression and purification of human estrogen-related receptor γ-ligand binding domain hERRγ-LBD recombinant protein

[0045] The total RNA of human colorectal adenocarcinoma cell Caco-2 was extracted with Trizol reagent, and reversed into cDNA as a template. Referring to the coding sequence of human estrogen-related receptor ERRγ (NCBI GenBank: NP_001230435.1) published by NCBI, primers were designed to amplify its sequence. Using the double enzyme digestion method, the coding gene of hERRγ-LBD (44..263aa) was connected to the pGEX-4T-1 vector, transformed into E.coli DH5α competent cells, and the positive clones were screened and sequenced to obtain the recombinant expression vector pGEX-4T - hERRγ-LBD. The recombinant plasmid was extracted, transformed into E.coli BL21 competent cells, and hERRγ-LBD recombinant expression bacteria were obtained. The recombinant bacteria were amplified and cultured at 37°C, and the expression was induced by adding IPT...

Abstract

The invention discloses a kit for rapidly detecting endocrine disturbance activity of chemicals. The kit comprises a recombinant human nuclear receptor ligand binding domain hNR-LBD protein solution, recombinant human nuclear receptor co-activation factor-EcoR I fusion protein solution, a specific Taqman probe, A-DTT buffer solution and B buffer solution. The invention further discloses a method for rapidly detecting the endocrine disturbance activity of the chemicals. According to the method, the endocrine disturbance activity of the chemicals can be rapidly detected on a microwell plate by applying the kit. Through the Taqman fluorescence probe method, the high-flux, multi-target and rapid detection of the endocrine interference activity of chemicals such as alkylphenol, p-alkoxy phenol, nipagin esters, bisphenol, biphenyl, diphenyl ethers, organophosphorus and halogen compounds is realized, and the upper limit of detection reaches 10-11M.

Description

technical field [0001] The invention relates to the technical field of detection of environmental compounds, in particular to a kit and method for rapidly detecting the endocrine disrupting activity of chemicals based on fluorescent probes. Background technique [0002] There are hundreds of thousands of newly synthesized chemicals in the world every year. According to the statistics of Chemical Abstracts Service (CAS), there are more than 120 million registered compounds, and the number is still increasing. Endocrine Disrupting Chemicals (EDCs) refer to environmental compounds that can interfere with the synthesis, secretion, transport, metabolism, binding and removal of endogenous hormones to affect the homeostasis, reproduction and development of organisms. Studies have shown that many chemicals in the environment have endocrine disrupting effects. They come from a wide range of sources and have greatly different structural properties. There are persistent organic polluta...
