Embodiment 1

[0025] 1. Seed collection: collect mature fruit clips, process the outer testa to obtain yellow-brown water shield seeds, and set aside.

[0026] 2. Seed pretreatment: soak the seeds selected in step 1 with 75% alcohol for 5 minutes, and rinse them with tap water. Then disinfect with 2% sodium hypochlorite solution for 20 minutes, and rinse with sterile water 3 times.

[0027] 3. Preparation of soaking solution: take gibberellin GA 3 Dilute to 1.0, 1.5, 2.0 and 2.5mmol / L with water respectively.

[0028] The pretreated seeds were subjected to the following two treatments:

[0029] 1) Gibberellin soaking treatment after cold-wet stratification

[0030] The pretreated Brasenia seeds were subjected to cold-wet stratification, wherein the seed: wet perlite volume ratio ≈ 1:3 and mixed well. Store in a refrigerator at 4°C for 0-240 days to ensure humidity. Afterwards, 50 treated seeds were taken as one portion, each group was repeated 4 times, soaked in 50mL of 0, 1.0, 1.5, 2....

Embodiment 2

[0041] 1. Seed collection: collect mature fruit clips, process the outer testa to obtain yellow-brown water shield seeds, and set aside.

[0042] 2. Seed pretreatment: soak the seeds selected in step 1 with 75% alcohol for 5 minutes, and rinse them with tap water. Then disinfect with 1:20 sodium hypochlorite solution for 20 minutes, and rinse with sterile water 3 times.

[0043] 3. Rapid dehydration of seeds with silica gel for 0, 1, 3, 6, 12, 18, 24, 30 and 36 hours, wherein the volume ratio of seeds to silica gel is ≈1:3 and the final water content is 24.0% and 18.1% respectively , 16.0%, 13.1%, 9.7%, 8.2%, 7.2%, 6.7% and 6.1%.

[0044] 4. The dehydrated seeds are subjected to cold-wet stratification treatment, in which the volume ratio of wet perlite is ≈1:3, mixed evenly, and stored in a refrigerator at 4°C for 0-135d to ensure humidity. The wet perlite in this embodiment is obtained by uniformly mixing 1 part by weight of perlite and 1.4-2 parts by weight of water.

Abstract

The invention belongs to the technical field of germination of plant seeds and particularly relates to a method to improve germination of Brasenia schreberi seeds and application of the method. Dormancy breaking and germination of Brasenia schreberi seeds are promoted herein through cold-damp stratification. Gibberellin (GA3) soaking of seeds after cold-damp stratification helps increase the germination rate of the Brasenia schreberi seeds; especially 240 days after cold-damp stratification, the Brasenia schreberi seeds are treated with 2.5 mmol / L gibberellin solution for 6 hours, germinationrate of the Brasenia schreberi seeds is up to 76.0+ / -0.8%. The Brasenia schreberi seeds are normal seeds; suitable dehydration helps promote the dormancy breaking and quick germination of the Braseniaschreberi seeds; especially hydration is carried out until water content of the Brasenia schreberi seeds is 9.7+ / -0.1%, and germination rate of the Brasenia schreberi seeds reaches 84.0+ / -2.8% afterthe Brasenia schreberi seeds are subjected to cold-damp stratification for 135 days. The method herein is a current optimal scheme to rapidly increase the germination rate of fresh mature Brasenia schreberi seeds.

Description

technical field [0001] The invention belongs to the technical field of plant seed germination, and in particular relates to a method for improving water shield seed germination and an application thereof. Background technique [0002] Brasenia schreberi (Brasenia schreberi J.F.Gmel.) is a perennial aquatic herb of Brasenia genus (Cabombaceae). Produced in China, the Soviet Union, Japan, India, the United States, Canada, eastern Oceania, and western Africa. In my country, it is mainly distributed in areas south of 30° north latitude. Ponds, rivers, lakes, or swamps with good water quality are mainly concentrated in Jiangsu, Zhejiang, Jiangxi, Hubei, Hunan, Sichuan, and Yunnan. Water shield is a remnant and rare species of Tertiary flora, a world-renowned rare aquatic vegetable, has a long history of cultivation, and has high edible and medicinal value. [0003] For a long time, the main propagation method of water shield is stem cutting, that is, asexual reproduction; long-t...
