Abstract
The invention discloses an application of dexmedetomidine in preparation of a pharmaceutical composition for improving diabetic peripheral neuropathy, a rat type II diabetes model and a rat RSC96 cell line are prepared through a high-fat and high-glucose combined small-dose intraperitoneal injection streptozotocin (STZ) method, a DPN model is established in vitro, DEX, DEX and an alpha2 receptor blocker yohimbine are applied, the effects of DEX and a transfected miR-34a reinforcing agent mimics, DEX and a transfected miR-34a inhibitor antitagomir, DEX and a transfected SIRT1 overexpression plasmid, and DEX and a transfected SIRT1 silent plasmid on rats and cell lines are provided, the influence of DEX on DPN is judged by detecting cell apoptosis, and the regulation mechanism of improving diabetic rat peripheral neuropathy by regulating miR-34a in a targeting manner to regulate SIRT1 is discussed for the first time.
