By establishing the system matrix model of microscope imaging, various uncertain factors in the data collection process are brought into a unified description framework; based on the image sparse feature, the image axial stretching effect due to missing cone is effectively alleviated, and the axial resolution of a microscope is improved; and meanwhile, the phase data is acquired by sparse angles, the total acquisition angle quantity is reduced, the imaging time is reduced, the time resolution of the microscope is improved, and the observation of the dynamic characteristic of biological samples is facilitated.
