Abstract

The invention discloses a method for rapidly identifying a Fraxinus velutina and Fraxinus pennsylvanica hybrid by using SSR (simple sequence repeat) marking technology. Genome DNA of Fraxinus velutina and Fraxinus pennsylvanica parents and their hybrid is used as a template, and an SSR primer is designed by using a constructed Fraxinus velutina transcriptome database; SSR markers with high, stable and clear polymorphism and parent complementary bands are screened out, hybrid plants are identified to obtain a pair of suitable SSR markers, the markers amplify a special band identical with a female parent from hybrid offspring, and hybrids having the female parent special band are true hybrids. The method enables quick and accurate identification for the authenticity of Fraxinus velutina and Fraxinus pennsylvanica hybrids, and good basis is laid for identifying hybrids of different combinations of Fraxinus parents.
