Problems solved by technology

Scott A. Bergfeld et al pointed out in the review article Bone marrow-derived mesenchymal stem cells and the tumor microenvironment that there are still serious differences in the scientific community on whether mesenchymal stem cells promote tumor growth or inhibit tumor production. The role of tumor type is not clear

[0056] Example 2: Effects of Modified BMSCs on the Growth of Pancreatic Cancer Cells

[0057] The inventors studied the effect of bone marrow mesenchymal stem cells downregulated by miR-134 on the growth of pancreatic cancer PC-3, pancreatic cancer PANC-1, and pancreatic cancer SW1990, and found no promotion effect on the growth of pancreatic cancer cells.

[0058] The present invention confirms that after the low expression of miR-134 in mesenchymal stem cells, the tropism of mesenchymal stem cells to pancreatic cancer PC-3, pancreatic cancer PANC-1, and pancreatic cancer SW1990 are all enhanced, and the targeting is increased; miR-134 After low expression, the expression of FOXM1 gene was up-regulated, and the content of FOXM1 protein was significantly increased, which proves that miR-134 improves the tendency of mesenchymal stem cells to pancreatic cancer PC-3, pancreatic cancer PANC-1, and pancreatic cancer SW1990 by regulating its target gene FOXM1. sexual. Abstract

The invention discloses application of miR-134 and a target gene FOXM1 to preparation of a mesenchymal stem cell-based pancreatic cancer targeted vector. According to discovery of the invention, after low expression of the miR-134 of the mesenchymal stem cell, the tropisms of the mesenchymal stem cell on pancreatic cancer PC-3, pancreatic cancer PANC-1 and pancreatic cancer SW1990 are enhanced and the targeting proper is enhanced; and after miR-134 low expression, the FOXM1 gene expression is up-regulated, and the protein content of the FOXM1 is obviously increased, so the miR-134 is used for enhancing the tropisms of the mesenchymal stem cell on pancreatic cancer PC-3, pancreatic cancer PANC-1 and pancreatic cancer SW1990 by regulating the target gene FOXM1. A medicine for inhibiting miR-134 and up-regulating FOXM1 gene and protein expression can be used for improving the targeting property of the mesenchymal stem cell on the pancreatic cancer, the modified BMSC does not have the effect of promoting the growth of pancreatic cancer cells, and the mesenchymal stem cell-based pancreatic cancer targeted vector can be prepared.

Description

technical field [0001] The invention belongs to the field of stem cells, and relates to targeting tumor tissue using stem cells as a carrier, in particular to the application of miR-134 and its target gene FOXM1 in preparing a mesenchymal stem cell-based pancreatic cancer targeting carrier. Background technique [0002] Mesenchymal stem cells (mesenchymal stem cells, MSCs) have the ability of self-renewal and multiple differentiation potentials. A large number of experiments have used this characteristic of MSCs to study new therapeutic methods for related diseases. Gene therapy is considered to be one of the most promising options for treating tumors after traditional therapies such as surgery, radiotherapy and chemotherapy. The primary problem of gene therapy is to choose a suitable carrier, and the tumorigenicity of mesenchymal stem cells makes it possible to become an effective carrier for tumor gene therapy. [0003] On the targeting effect of mesenchymal stem cells on...
