Problems solved by technology

This method is a commonly used clinical method at present, and has the following defects: First, the growth potential of MoDCs formed in vitro is limited and it is difficult to culture for a long time. Therefore, multiple rounds of vaccination need to be repeatedly prepared. Second, the mature and activated MoDCs in vitro are not CD141+DCs in the true sense, and their functions cannot be compared with primary DCs
But so far, there is no report that Tax protein of non-human primate T lymphocyte virus can induce dendritic cells to mature and maintain high activity in vitro

Embodiment Construction

[0025] Unless defined otherwise, all technical and scientific terms used herein have the meaning commonly understood by one of ordinary skill in the art.

[0026] Through comparative analysis of the amino acid sequences of Tax proteins in HTLVs and STLVs viruses, the inventors found that the homology of Tax in PTLVs viruses is very high, reaching more than 74%. Among them, the homology between HTLV1 and STLV1Tax is as high as 95%, and the homology between HTLV3 and STLV3Tax is also as high as 90%. These information suggest that the function of Tax protein in STLVs virus may be similar to that of HTLV Tax protein, and the mature highly active dendritic cells may be generated through Tax protein of STLVs virus.

[0027] The amino acid sequences of STLV1, STLV2, STLV3, and STLV4 viral Tax proteins are shown in SEQ ID NO: 1-4, respectively. The present inventors isolated mononuclear cells from peripheral blood or umbilical cord blood, cultured them in the presence of cytokines, i...

Abstract

The invention provides a method for preparing mature high-activity dendritic cells. The method comprises the following steps of (1) collecting single karyocytes from peripheral blood or umbilical cordblood; (2) inducing the single karyocytes to be differentiated into dendritic cells; (3) enabling the dendritic cells to be in contact with the non-human primate animal T lymphotropic virus protein from non-pathogenic; (4) screening and identifying the obtained mature high-activity dendritic cells. The invention provides the mature high-activity dendritic cell prepared by the method, and a use method of the cell. The dendritic cell prepared by the method has strong dendritic cell reproductive capability and high activity; the gene modification transmission is convenient; the cell is superiorto the dendritic cell used in the prior art. The cell and the use method have the wide application prospects in aspects of tumor or virus immunotherapy.

Description

technical field [0001] The invention relates to a method for preparing dendritic cells, especially a method for preparing mature highly active dendritic cells in vitro. The present invention also relates to mature highly active dendritic cells prepared by the method and related applications thereof. Background technique [0002] The immune system is the most important system for the human body to recognize and remove foreign pathogenic microorganisms (viruses) and mutated cells (tumors) in the body. The functional units that play a role in the immune system are various types of immune cells, including macrophages, B lymphocytes, T lymphocytes, natural killer cells (NK) and dendritic cells. Among them, B lymphocytes resist the invasion of foreign antigens by producing antibodies and mainly participate in humoral immunity; T lymphocytes and NK cells mainly mediate cellular immunity. As antigen-presenting cells, dendritic cells play a key role in cellular immunity mediated by...
