Abstract

An arrangement for supplying immersion media. The arrangement includes a media supply unit for the controlled supply of a medium or of a mixture into a contact region between an optical lens and a specimen slide, on which a specimen may be arranged. An image capture unit is provided for capturing image data on the basis of detection radiation from the object space along a detection beam path extending through the contact region. An evaluation unit is provided to establish current image parameters on the basis of captured image data, to compare said current image parameters to intended image parameters and to establish a desired mixing ratio of at least two components of the medium depending on the comparison. A mixing apparatus for the controlled setting of the mixing ratio of the components of the medium to be introduced into the contact region and a control unit for actuating the mixing apparatus depending on the established desired mixing ratio are parts of the arrangement. In addition, the invention relates to a method for setting optical parameters of a medium.

Description

technical field [0001] The invention relates to an arrangement and a method for setting the optical properties of a medium, in particular an immersion medium, and to a microscope comprising such an arrangement. Background technique [0002] Especially in live cell microscopy, the precise refractive index of the sample is often not known. This does not (only) concern the embedding medium and / or the buffer employed, but more particularly the interior of the sample, which may be provided by eg cells, (tumor) spheroids, tissue sections or whole organisms. [0003] Because these samples are usually stored and examined in aqueous solutions (buffered solutions), water immersion objectives are often used - also for historical reasons. These are optimized for a refractive index of n = 1.33 (refractive index of water). However, the refractive index of living cell samples deviates from n = 1.33 (see for example, Liu, P.Y. et al. "Cell refractive index for cell biology and disease dia...
