[0074] For our study, we developed an experimental model to mimic COPD disease in mice by chronic exposure to cigarette smoke (CS) (13). These mice have increased susceptibility to respiratory pathogens associated with defective IL-22 production (14, 15). In contrast, these animals showed increased expression of IL-19, IL-20, and IL-24 in response to S. pneumoniae or NTHi. Treatment with an anti-IL-20Rb blocking antibody increased bacterial clearance in control mice infected with Streptococcus pneumoniae (12) and protected CS-exposed mice from bacterial infection by reducing bacterial load and inflammatory infiltration (WO2016083304) . In view of these results, we hypothesized that IL-20 cytokine might contribute to the increased susceptibility of COPD mice to bacterial infection through its inhibitory effect on Th17 cytokine production.

Abstract

Chronic obstructive pulmonary disease (COPD) is still the major cause of global morbidity and mortality. A COPD acute exacerbation patient (AE-COPD) is mainly caused by respiratory tract infection and is related to inevitable decline of lung function, edema exacerbation and airway and systemic inflammation. Previous results show that treatment with an anti-IL-20Rb blocking antibody increases bacterial clearance in control mice infected with Streptococcus pneumoniae and protects CS exposed mice from bacterial infection by reducing bacterial load and inflammatory infiltration. Therefore, people are of interest in the production of monoclonal antibodies specific for IL-20Rb with neutralizing activity for AE-COPD treatment. This need is met by providing an antibody that is specific for IL-20Rb.
