Problems solved by technology

[0005] At present, most of the studies on Toxoplasma infection of wild animals such as civet cats are focused on epidemiology. Serological methods are used to detect Toxoplasma gondii antibodies, PCR methods are used to detect Toxoplasma gondii DNA in brain tissue, and pathological tissue sections are used to observe Toxoplasma gondii infection causes tissue and cellular damage, but there is no report on the application of transcriptome sequencing technology to study the differentially expressed genes in civet brain tissues infected by Toxoplasma gondii

[0050] 1 Materials and methods

[0051] 1.1 Materials

[0052] Clean-grade civet cats aged 3 to 8 months were purchased from Jiahe Special Breeding Base in Shaoguan, Guangdong Province; Toxoplasma gondii ME49 strains were preserved by the Parasite Laboratory of Veterinary College of South China Agricultural University.

[0053] 1.2 Sample preparation

[0054] Establishment of Toxoplasma gondii-infected civet model

[0055] Twelve 4-month-old male civet cats of SPF grade were purchased from Jiahe Special Breeding Base in Shaoguan, Guangdong Province, and 6 of them were marked as the experimental group and the other 6 as the control group. In the experimental group, the cysts of the Toxoplasma gondii strain ME49 were injected into the mice by intragastric administration, the number of cysts was 40 per mouse, and they were diluted to 5 mL per mouse with normal saline...

Abstract

The invention provides a screening method of a toxoplasma gondii infected animal host brain tissue differential expression gene and application thereof, and the gene expression difference condition of the toxoplasma gondii infected animal host brain tissue is analyzed by using a transcriptome sequencing technology in combination with bioinformatics; and screening and identifying differential expression genes of the brain tissue after the toxoplasma gondii infects the animal host. The method can be used for quickly and effectively researching gene expression difference, reference is provided for revealing body immune response after an animal host is infected with the toxoplasma gondii, 10 differential expression genes CCL5, CCL8, CCL23, CASP8, TLR1, TLR4, STAT1, ROS1, NOS1 and NOS2 in the animal host are verified by adopting a qRT-PCR method, the screened differential expression genes are used as target genes for diagnosing brain infection of the host toxoplasma gondii, and the method has the advantages that the research on the gene expression difference is realized, and the application prospect is wide. The method is beneficial to accurate discrimination of toxoplasma gondii infection of animal host brain including paguma larvata and other wild animals, and provides a diagnostic target for diagnosis of toxoplasma gondii infection of wild animals.

Description

technical field [0001] The invention relates to the field of parasitology, in particular to a method for screening genes differentially expressed in the brain tissue of animal hosts infected by Toxoplasma gondii and its application. Background technique [0002] Toxoplasma gondii is a protozoan obligately parasitic in eukaryotic cells. Toxoplasmosis caused by Toxoplasma gondii infection is an important zoonotic disease, which can lead to severe clinical symptoms and even death in infants, pregnant women and immunocompromised persons. Humans mistakenly eat food or water containing Toxoplasma oocysts It is one of the main ways to cause toxoplasmosis. Toxoplasma has three infective stages: sporozoites, tachyzoites, and bradyzoites. Toxoplasma gondii tachyzoites can widely invade almost all nucleated cells, and favor neurocytes, which can lead to toxoplasma gondii encephalitis. Epidemiological surveys show that about 30% of the world's population has potential Toxoplasma infe...
