A single-use bioreactor or disposable bioreactor is a bioreactor with a disposable bag instead of a culture vessel. Typically, this refers to a bioreactor in which the lining in contact with the cell culture will be plastic, and this lining is encased within a more permanent structure (typically, either a rocker or a cuboid or cylindrical steel support). Commercial single-use bioreactors have been available since the end of the 1990s and are now made by several well-known producers (See below) .

A bioreactor includes a first shell a first outer membrane and the first growth membrane. The first shell includes a first sidewall and a first end wall. The first sidewall defines a first chamber and has a first open end and a second open end that is opposite the first open end. The first end wall extends across the first open end of the first sidewall. The second shell includes a second sidewall that defines a second chamber. The first outer membrane is secured to the first end wall and is configured to substantially seal the first chamber at the first end wall. The first growth membrane is disposed in the second chamber. The first growth membrane is microporous and is configured to allow liquid media to pass through while retaining cells with in the second chamber.

A multiport device for connecting a loop, preferably a tangential flow filtration loop or a sensor loop, to one port of a bioreactor, preferably a single-use bioreactor, is configured to be fixed to the port of the bioreactor. The multiport device includes a first flow path configured for withdrawing fluid from the bioreactor, and a second flow path configured for supplying fluid to the bioreactor. The first flow path has a first end adapted to be in fluid connection with the bioreactor, and a second end adapted to be connected to an inlet of the loop. The second flow path has an outer end adapted to be connected to an outlet of the loop, and a mouth adapted to be in fluid connection with the bioreactor. The mouth of the second flow path is distanced from the first end of the first flow path by at least 5 mm, preferably 10 mm.
