Embodiment Construction

[0035] 1. Experimental animals

[0036] The zebrafish used in the experiment were all Tübingen strains, purchased from the State Key Laboratory of Medical Genomics, Shanghai Second Medical University. Breeding, breeding, mating and egg collection were carried out according to the method in the zebrafish manual. The water temperature was 28.5°C, the light / dark light cycle was 14h / 10h, the embryo culture medium was Holtfreter buffer, and some embryos were treated with 0.003% PTU to prevent pigment formation.

[0037] 2. Antisense suppression technology and overexpression technology

[0038] In order to obtain the Loss of Function phenotype in miRNA functional studies, we use the antisense inhibition technology of morpholinos oligonucleotides. Serial MOs, standard negative control morpholino (MO-NC) and p53 MOs were purchased from Gene Tools, and each package was 300nmol. In addition to MO-219, miR-34a, miR-9*, miR-146b, miR-181a, miR-92b, miR-125b, miR-129, miR-130a,...

Abstract

The invention belongs to the field of biotechnology and medicine and relates to an application of miR-219, a new marker of human brain glioma. The fact that the expressions of miR-219 in the human brain glioma tissues and cells are obviously reduced is confirmed for the first time, and the facts that miR-219 can directly control Hh-Gli2 and Pdgfra signal paths during early development of the neural tube of embryo and occurrence of the brain glioma and can obviously suppress proliferation and infiltration of human glioma cells are confirmed. Further quantitative analysis of clinical brain glioma specimens at different levels shows that miR-219 can serve as the marker for detecting the brain glioma.

Description

technical field [0001] The invention relates to the use of small RNA molecules, in particular to the use of miRNA-219, and belongs to the technical field of biomedical materials. Background technique [0002] Recent studies have shown that miRNAs can function as tumor suppressor genes or oncogenes, and their mutation or ectopic expression is closely related to human cancer. Glioma is the most frequent brain malignancy in adults. Studies have shown that dysregulation of some miRNAs (such as miR-21, miR-10b, miR-221 / 222, miR-26a, miR-124, miR-128 and miR-34a) in glioma can affect tumor cell proliferation , apoptosis, invasion, metastasis and angiogenesis. Finding miRNA target genes and revealing their mechanism of action is not only one of the problems to be solved in the post-genomic era, but also is expected to provide new strategies for the diagnosis and treatment of tumors and other diseases. [0003] miRNAs are extremely abundantly expressed in the brain, but the inter...
