Problems solved by technology

[0007] However, what has been known so far is limited to the production of cellobiohydrolases belonging to GH family-6 and GH family-7 (for example, Non-Patent Document 1) and four types of enzymes belonging to GH family-5 and GH family-12. endoglucanase (for example, non-patent literature 2, 3), but no endoglucanase belonging to GH family-45 has been found, and the detailed cellulolytic mechanism has not been elucidated

[0094] (Preparation method of protein with endoglucanase activity)

[0095] The protein having endoglucanase activity of the present invention can be obtained from a culture of Basidiomycetes, and preferably, can also be prepared according to a method using the transformant of the present invention.

[0096]

[0097] The method for preparing a protein with endoglucanase activity using basidiomycetes at least includes a process for culturing basidiomycetes (culture process) and a process for extracting proteins with endoglucanase activity from the culture (extraction Process), further including other processes as needed. This preparation method is the first embodiment of preparing the protein with endoglucanase activity of the present invention.

[0098] -Cultivation process-

[0099] The culture process is a process of culturing Basidiomycetes.

[0100] The basidiomycetes are not particularly limited and can be appropriately selected according to the purpose, but are preferably basidi...

Embodiment

[0203] Hereinafter, the present invention will be described in further detail with reference to the embodiments, but the present invention is not limited to these embodiments.

[0204] (Exploration of genes for novel proteins with endoglucanase activity)

[0205] In order to obtain the gene of a novel protein with endoglucanase activity, first, by the following method, for the Basidiomycete Phanerochaete chrysosporium (Phanerochaete chrysosporium) K-3 strain (Johnsrud, SC and KEEriksson.1985. Cross breeding of selected and mutatedhomokaryotic strains of Phanerochaete chrysosporium K-3-New cellulasedeficient strains with increased ability to degrade lignin.Appl.Microbiol.Biotechnol.21:320-327.) was studied.

[0206] The above-mentioned Phanerochaete chrysosporium K-3 strain was added to Kremer and Wood medium (Kremer, SM and PM Wood. 1992. Evidence that cellobiose oxidase) containing 2% cellulose (CF11; manufactured by Whatman). from Phanerochaetechrysosporium is primarily an Fe(III)...

Abstract

Disclosed are: a novel protein having an endoglucanase activity, which is found in a basidiomycete (into which edible mushrooms are classified) and is user-friendly from a safety standpoint and the like; novel DNA encoding the protein; a vector carrying the DNA; a transformant harboring the vector; a method for producing a protein having an endoglucanase activity by using the transformant; a method for producing a sugar by using at least one member selected from a protein having an endoglucanase activity and a basidiomycete; a method for producing ethanol by using a sugar produced by the aforementioned method; a food, a feed and a detergent each comprising a protein having an endoglucanase activity; and a method for treating a cellulose-containing woven fabric by using a protein having an endoglucanase activity. The protein is characterized by being derived from a basidiomycete, having a molecular weight of 18 kDa as determined by SDS-PAGE, and having an endoglucanase activity.

Description

Technical field [0001] The present invention relates to a novel protein with endoglucanase activity, a novel DNA encoding the protein, a vector containing the DNA, a transformant containing the vector, and the preparation of the transformant with endoglucanase activity The method of protein, and the method of preparing sugar using at least any one of the protein with endoglucanase activity and basidiomycetes, the method of preparing ethanol using the sugar obtained by the sugar preparation method, and the method of preparing ethanol with Foods, feeds, detergents with proteins with endoglucanase activity, and methods for treating cellulose-containing fabrics with proteins with endoglucanase activity. Background technique [0002] Endoglucanase (EGs (endoglucanase); EC 3.2.1.4) is one of the enzymes that hydrolyze cellulose. Enzymes that hydrolyze cellulose also include cellobiohydrolase (CBHs (cellobiohydrolase); EC3.4.1.91), β-glucosidase (BGLs (beta-glucosidase); EC 3.2.1.21), ...
