Embodiment 1

[0060] Example 1. Auxiliary screening of the main QTL locus (QPhs.caas-2BL) and the acquisition of special primer pairs for high panicle-resistant germinating wheat

[0061] 1. Identification of panicle germination resistance

[0062] The hybrid offspring F of parent CA0431 resistant to panicle germination and parent CA0306 susceptible to panicle germination 2 A total of 220 plants were planted in Beijing in 2010-2011 in the generation group, with a row length of 2m and 20 seeds / row.

[0063] From June to July 2011, F in Beijing 2 The identification of ear germination resistance of the generation population, the specific method is as follows: F 2 For the generation population, 5 ears were taken from each plant, and samples were taken at the physiological maturity stage. They were first placed in a ventilated place to dry naturally for 5 days, and then placed in a refrigerator at -20°C to maintain dormancy. After all samples were taken, they were air-dried for 2 days, and the...

Abstract

The invention discloses a method and a special primer for screening or auxiliary screening of wheat with high pre-harvest sprouting resistance. The special primer for screening or auxiliary screening of the wheat with high pre-harvest sprouting resistance consists of a primer pair 1 and a primer pair 2 as follows: the primer pair 1 consists of single-stranded DNA molecules shown in sequence 1 in a sequence table and single-stranded DNA molecules shown in sequence 2 in the sequence table, and the primer pair 2 consists of single-stranded DNA molecules shown in sequence 3 in the sequence table and single-stranded DNA molecules shown in sequence 4 in the sequence table. According to the invention, the experiment proves that the invention provides the special primer which utilizes an offspring group produced by CA0431 / CA0306 for QTL (Quantitative Trait Locus) mapping to find out a molecular marker QTL (QPhs.caas-2BL) which is closely linked with a CA0431 pre-harvest sprouting resistance gene, and screens out the special primer for auxiliary screening of the wheat with the pre-harvest sprouting resistance based on the main effect QTL. The special primer can be utilized for screening or auxiliary screening of the wheat with pre-harvest sprouting resistance so as to cultivate pre-harvest sprouting resistant varieties.

Description

technical field [0001] The invention relates to the field of biotechnology, in particular to a method for screening or assisting in screening high panicle-resistant germination wheat and special primers thereof. Background technique [0002] Ear germination (Pre-harvest sprouting referred to as PHS) means that wheat encounters continuous rainy weather or germinates on ears in a humid environment before harvest. Spike germination not only affects yield, but also seriously affects quality (especially processing quality), storage, and next year's sowing quality, causing different economic losses. [0003] Many countries and regions in the world have been harmed by ear germination, among which Canada and Australia are the most serious. According to Derera reports, an average of 7% of the wheat in Australia is damaged by ear germination every year. In 1984, the most serious year, the degree of ear germination damage was as high as 20%. On average, Canada loses nearly $100 milli...
