Embodiment 1

[0074] Embodiment 1, the preparation of the collagen support of repairing peripheral nerve injury

[0075] 1. Preparation of Collagen Scaffolds Containing Dual Factors for Functional Repair of Peripheral Nerve Injury

[0076] 1. Preparation of Collagen Tubes

[0077] According to the radius of the nerve injury site to be repaired, find a mold with the same radius, choose glass or plastic as the mold, cut the collagen film into an appropriate size, soak it in triple distilled water for 2 minutes to make the collagen film soft, and wrap the collagen film around the mold Wrap 5 / 4 circles, wrap it with liquid collagen, and let it dry overnight ( figure 1 Middle A). room temperature (25 ℃ overnight (12 hours) cross-linking, after cross-linking, the collagen tube was removed from the mold, lyophilized, and irradiated for later use ( figure 1 Middle B and C).

Embodiment 2

[0094] Example 2. Application of Peripheral Nerve Injury Function Repair Collagen Scaffold to Treat Long-distance Transverse Nerve Injury in Minipigs

[0095] 1. Prepare a long-distance (3.5cm) facial nerve transection injury model in minipigs, and bridge the peripheral nerve injury function repair collagen scaffold material at both ends of the nerve injury

[0096] First, a model of facial nerve transection injury in minipigs was prepared. Select 8 13-month-old miniature pigs (China Agricultural University), with a body weight of about 40-45 kg. After muscle anesthesia, the facial nerves on both sides of the pigs were cut free by 3.5 cm to make a long-distance facial nerve injury model of miniature pigs ( figure 2 Middle A).

[0097] Then, 6 of the miniature pigs were sutured to bridge the four kinds of collagen scaffolds prepared in Example 1 at the proximal end and the distal end of the nerve injury ( figure 2 In middle B), three sets of replicates were set up for each ...

Abstract

The invention discloses a method for preparing a collagen material for repairing peripheral nerve injury. The method comprises the following step: co-incubating a ciliary neurotrophic factor with a collagen binding domain (CBD-CNTF), a basic fibroblast growth factor with a collagen binding domain (CBD-bFGF) and an orderly collagen material to obtain a collagen material for repairing the peripheral nerve injury. According to the preparation method, two factors CBD-CNTF and CBD-bFGF are simultaneously crosslinked to the orderly collagen scaffold material, and the action of the double-factor peripheral nerve injury repairing collagen material in long-cross-section nerve injury repairing can be evaluated by establishing an animal peripheral nerve-facial nerve long cross-section injury model. Results prove that compared with single factor CBD-CNTF or CBD-bFGF functional collagen scaffold material, the double-factor peripheral nerve injury repairing collagen material can be used for better promoting regeneration of facial nerves and restoration of functions, and has a more important realistic guiding magnificence to application of clinical peripheral nerve injury repairing in the future.

Description

technical field [0001] The invention belongs to the field of medical materials, and relates to a collagen material for repairing peripheral nerve damage, in particular to a collagen material for repairing peripheral nerve damage containing two factors and a preparation method thereof. Background technique [0002] Peripheral nerve injury is one of the common diseases in clinical practice. The current clinical application of direct stump suture technique or autologous nerve transplantation cannot fully solve the clinical difficulties due to its application limitations. With the development of tissue engineering, the use of tissue engineering methods to promote nerve regeneration to repair peripheral nerve injury has become a research hotspot. [0003] Collagen scaffold material can bridge the growth of axons and play a role in guiding the growth of axons. Collagen is a biomaterial widely used in tissue engineering because of its low immunogenicity, good biocompatibility and ...
