The circulating tumor cell separating and enriching device is characterized by comprising an acquisition channel, an anticoagulation channel, a control valve, a blood collector, a blood collecting bag, a CTC acquisition filter and a blood transfusion channel; the acquisition channel and the anticoagulation channel are respectively or jointly connected to an input end of the control valve, a switching end of the control valve is connected with an input end of the blood collecting bag, the blood collecting bag is arranged on the blood collector at a blood acquisition phase, an output end of the blood collecting bag is sequentially connected with the CTC acquisition filter and the blood transfusion channel, and an output end of the blood transfusion channel is connected with another switching end of the control valve.
