Embodiment Construction

[0029] Cloning, identification and functional analysis of the full-length sequence of a novel cotton EIN3 gene GhEIN3.

[0030] 1. Isolation and cloning of GhEIN3 gene and cDNA sequence

[0031] More than 10,000 cotton cDNAs were isolated from the cotton fiber cDNA library. Through bioinformatics analysis, a full-length cDNA sequence of a GhEIN3 gene was identified. The sequence is shown in SEQ.ID.No.1, and the gene coding region is the first (from 194–2035bp, underlined as 5'-untranslated region 193bp (5'-UTR, from 1–193bp) and 3'-untranslated region 282bp (3'-UTR, from 2036–2317bp); the gene contains 1842bp Open reading frame (ORF), encoding a protein of 613 amino acids, as shown in SEQ.ID.No.2, the gene molecular weight 69.40KD.The N-terminus of GhEIN3 protein contains a conserved EIN3 superfamily domain, and Arabidopsis thaliana AtEIN3 and AtEIL1 protein homology are 64% and 64.5% respectively. With GhEIN3 open reading frame sequence design primer GhEIN3OEup and GhEIN3OEdn...

Abstract

The invention relates to authentication and application of a novel cotton fiber development relevant gene GhEIN3. The gene does not contain intron, wherein the cDNA full-length sequence of the gene is 2317 bp, and the gene contains 5' end untranslated region 193 bp, an open reading frame 1842 bp and 3' end untranslated region 282 bp, and protein containing 613 amino acids is encoded and is then advantageously expressed in the initial development period of cotton fiber. The GhEIN3 protein has transcription self-activation capacity and positioned in a cell nucleus, and it is shown that the protein plays a role of a transcription activation factor. GhEIN3 can form homodimer or heterodimer, and cotton fiber development is adjusted and controlled through mediate JA and GA hormonal signals. The number of GhEIN3 RNAi transgenic cotton short fiber (flock) is remarkably reduced, the number of long fiber (shag) is also reduced, the length of mature fiber (shag) is remarkably increased, the length of cotton fiber is decreased due to the overexpressed GhEIN3, and it is shown that GhEIN3 plays an important role in adjusting and controlling the cotton fiber initial and fiber elongation process.

Description

[0001] Technical field: [0002] The invention relates to a cotton gene, in particular to the cloning and functional identification of an EIN3 (Ethylene Insensitive 3) transcription factor gene GhEIN3 regulated by cotton fiber cell initiation and initiation elongation conversion. [0003] Background technique: [0004] Cotton is an important economic crop in my country, which is closely related to the income of 200 million farmers and the employment of 19 million textile workers in China. With the rapid development of the textile industry and the continuous improvement of people's living standards, the textile industry and its related industries have higher and higher requirements for the quality of cotton fibers. my country's total lint production ranks first in the world, and its per unit yield also ranks first among the five major cotton-producing countries. However, the quality of my country's cotton fibers is poor and cannot be used as raw materials for the production of s...
