This invention relates to the application of a nitrogen-efficient gene in maize, ZmLTPg14-NPF5.14. The nitrogen-efficient gene ZmNPF5.14 described in this invention promotes maize resistance to low nitrogen stress. Knocking out this gene reduces maize's resistance to low nitrogen stress. Mutant maize plants with this gene knockout exhibit more yellowing leaves under low nitrogen conditions compared to control plants, showing a more sensitive phenotype. ZmLTPg14 reduces maize's resistance to low nitrogen stress; mutant maize plants with this gene knockout are less sensitive to low nitrogen than control plants. Correspondingly, plants overexpressing ZmLTPg14 are more sensitive to low nitrogen. Furthermore, double mutants with both ZmLTPg14 and ZmNPF5.14 knockouts exhibit the same phenotype under low nitrogen conditions as those with ZmNPF5.14. The mutant of ZmLTPg14 can be used to cultivate maize varieties that are not sensitive to low nitrogen. The mutant plants of ZmNPF5.14, the overexpression plants of ZmLTPg14, and the double mutants of the two genes ZmLTPg14 and ZmNPF5.14 can be used to cultivate maize varieties that are sensitive to low nitrogen.

High-temperature-resistant methanotroph with high glutamic acid production and construction method and application thereof

The application discloses high-temperature-resistant engineered methanotrophs with high glutamic acid yield, a construction method and application thereof, and belongs to the field of industrial biotechnology. Methylotuvimicrobium buryatense 5GB1 as the host strain, transcriptional inhibition of the SOS response system by LexA protein is removed by knocking out a lexA gene in the genome of the host strain, expression of DNA damage repair related genes in the cell is activated, and high-temperature-resistant engineered methanotrophs MBΔ lexA that can stably grow in a high-temperature environment of 40-45 DEG C are obtained. The strain can still maintain normal growth and metabolism with methane as the sole carbon source under the condition of 40-45 DEG C, and the glutamic acid synthesis capacity of the strain is significantly better than that of the original strain at normal temperature. The high-temperature-resistant engineered methanotrophs MBΔ lexA provide important strain resources for the production of amino acid products by methanotroph cell factories in a high-temperature industrial scene, and are expected to improve the economy and sustainability of methanotrophic bioconversion.

Enterotoxigenic escherichia coli strain expressing f5, f41 pilus and application thereof

The application belongs to the field of microbiology and veterinary microbiological technology, and relates to a strain of enterotoxigenic Escherichia coli carrying F5 and F41 fimbriae and application thereof. The strain is isolated from a fecal sample of a diarrheal calf, and is confirmed to be enterotoxigenic Escherichia coli carrying F5 fimbriae-related genes, F41 fimbriae-related genes and ST enterotoxin estA gene through isolation and culture, morphological identification, biochemical identification, fimbriae gene and virulence gene PCR detection and whole genome sequencing analysis. The strain has been preserved in the China General Microbiological Culture Collection Center on June 13, 2022, and the preservation number is CGMCC No. 25077. Pathogenicity tests show that the strain has strong virulence and can stably establish an animal infection model. Drug sensitivity tests show that the strain has a specific sensitivity spectrum to a variety of antibacterial drugs. The strain can be used for animal infection model establishment, antibacterial drug evaluation, calf diarrhea pathogenesis research, prevention and control technology research, and can also be used for preparing biological agents for preventing or diagnosing calf diarrhea.

Under preferred conditions, the removal rate of nitrate nitrogen can reach 93.8%, and stable carbonate carbon fixation effect is realized.
