Abstract

The invention discloses a real-time fluorescence PCR identification method, primer and probe of a tiger derived component. The method comprises the following steps: 1, extraction of DNA from a sample; 2, real-time fluorescence PCR amplification detection; and 3, result determination. The primer comprises an upstream primer and a downstream primer, the sequence of the upstream primer is 5'-CTTTATCTGTCTATACATGCAT-3', and the sequence of the downstream primer is 5'-TGTAGGAGCCGTAGTATATT-3'; and the sequence of the probe is 5'-FAM-ACGAGGAATATACTACGGCT-Eclipse-3'. Compared with common PCR methods, the method disclosed in the invention has the advantages of simplicity in operation, and high efficiency and accuracy in identification, can effectively identify tiger night soil, tiger hair and tiger bones, and can be used for field tracing investigation and protection of tigers. The primer and the probe have very high specificity and sensitivity, and can be used for protecting and researching types of tigers.
