Embodiment Construction

[0020] The substantive content of the present invention will be specifically introduced below in conjunction with the embodiments. Due to space reasons, the description of the experimental process cannot be done in great detail. All the parts not described in detail in the experiment are routine operations well known to those skilled in the art.

[0021] 1. Experimental materials

[0022] The human hepatocyte tumor cell line HHCC was purchased from ATCC in the United States and preserved by our company. HHCC cells were resuscitated and inoculated into culture flasks, RPMI-1640 containing 10% FBS was added, and adherent cells were digested with 0.25% trypsin. Cells in the logarithmic growth phase were used for experiments. Balb / c mice were purchased from Nanjing Junke Bioengineering Co., Ltd.

[0023] The exogenous peptide of the present invention is synthesized by the standard polypeptide solid-phase synthesis technique known in the art, and adopts the N-terminal protection ...

Abstract

The invention discloses a gene target capable of improving anti-hepatoma effect of a DC vaccine, an inhibitor and a DC tumor vaccine. The invention proves that in vivo tumor suppression effect of the DC vaccine can be obviously improved by inhibiting expression of SOCS2 in DC, and a foreign peptide provided by the invention is an effective inhibitor for the expression of SOCS2 in DC, has a function similar to siRNA and can obviously improve in vivo tumor suppression effect of the DC vaccine. The SOCS2 expression inhibitor and the foreign peptide provided by the invention can be used for preparing a DC hepatoma vaccine with better anti-hepatoma effect.

Description

technical field [0001] The invention belongs to the field of gene therapy and immunotherapy, and in particular relates to a gene target, an inhibitor and a DC tumor vaccine for improving the effect of DC vaccine against liver cancer. Background technique [0002] Hepatocellular carcinoma is one of the most common malignant tumors worldwide, with approximately 600,000 patients diagnosed each year. At present, the treatment for HCC mainly includes surgical resection, liver transplantation, radiotherapy and chemotherapy, etc., but only 20% of the patients can be treated. The annual mortality rate of hepatocellular carcinoma in my country ranks second in cancer mortality. The etiology and pathogenesis of hepatocellular carcinoma have not yet been determined. Therefore, finding new therapeutic targets and drugs for hepatocellular carcinoma is a research focus. [0003] DC-based biotherapeutics are particularly attractive. Theoretically, DCs are the most effective APCs to stimu...
