[0051] The blood samples of 32 Landrace pigs from Hebei pig farms were collected respectively. After the samples were collected, they were stored in 75% alcohol and stored at -20°C. The genomic DNA of blood samples was extracted by phenol-chloroform extraction, and the specific steps were as follows:

[0052] (1) Preparation of ear tissue samples: Cut ear tissue samples of moderate size, wash away impurities and blood stains on the surface with normal saline, put them in a 1.5ml centrifuge tube, cut them into pieces with ophthalmic scissors, and add 500uL of tissue to it Lysis solution (Beijing Baitaike Biotechnology Co., Ltd.; product number: AU19011);

Abstract

The invention discloses a molecular marker for detecting the day-age for achieving a 100 kg pig based on a MEG3 gene, and applications thereof. The method for detecting the day-age for achieving a 100 kg pig by using the molecular marker is the following method (1) or (2): (1) detecting whether the 460 site deoxyribonucleotide of the second exon of the MEG3 gene of an individual pig is T or A or T and A; and (2) detecting whether the 847 site deoxyribonucleotide of the second exon of the MEG3 gene of an individual pig is A or G or A and G. According to the present invention, the experimental results prove that the determination result of the method for identifying the day-age for achieving the 100 kg pig is consistent with the actual determination result so as to provide the important significance for the selection of the excellent pig breeder.

Description

technical field [0001] The invention belongs to the field of genetic engineering, and in particular relates to a molecular marker for detecting pigs with a body weight of 100 kg and an application thereof based on the MEG3 gene. Background technique [0002] Pig is the first of the six livestock, and the pig industry occupies an important position in the world, especially in China's national economy. The proportion of pork in many meat products has been maintained at about one-third, and it is one of people's favorite non-staple foods. People's lives are constantly improving, and traditional breeding techniques can no longer meet people's demand for pork. With the development of modern breeding technology, the production performance of pigs has been significantly improved, and the healthy breeding and fattening of pigs is particularly important. [0003] The age of pigs reaching 100kg body weight is an important indicator for evaluating the growth rate of pigs, which can i...
