Embodiment 1

[0051] (1) Synthesis of colloidal gold

[0052] Add 100mL 1mmol / L HAuCl 4 The aqueous solution was added to the round bottom flask, the condenser tube was connected and reflux was started. Under magnetic stirring, heat to boiling, and quickly add 10mL of newly prepared 38.8mmol / L trisodium citrate solution. With the stirring and heating, the color of the solution changes from yellow to black and then gradually turns to dark red. The reaction is 15min. Afterwards, cool to room temperature naturally at room temperature and store at 4°C for later use.

[0053] (2) Preparation of colloidal gold probe

[0054] ① Activation of bio-barcode: The bio-barcodes labeled with sulfhydryl and 6-FAM, Cy3, and Texas red were centrifuged at 12000 rpm for 1 min according to the instructions, dissolved in TE buffer, and then TCEP (20 mmol / L) solution was added to make TCEP and DNA The ratio is 200:1, shaking at room temperature for 2-3h.

[0055] ② Antibody labeling: After the colloidal gold solution h...

Abstract

The invention provides an immunoassay kit for detecting poly-residue of organophosphorus pesticides and belongs to the technical field of pesticide detection. The kit includes: a black ELISA plate coated with an antigen, a colloidal gold probe solution, a washing solution, and a dithiothreitol solution. The antigen includes triazophos, parathion and chlorpyrifos; the colloidal gold probe is gold nano-particles, of which the surfaces are modified with bio-barcodes and any one of the specific monoclonal antibodies as follows: a triazophos antibody, a parathion antibody and a chlorpyrifos antibody; and the number of the bio-barcodes modifying the surfaces of the gold nano-particles is 130-170. The bio-barcodes are labeled with fluorophores, wherein the monoclonal antibodies and the colors ofthe fluorophores have a one-to-one corresponding relationship. The kit can quantitatively detect a plurality of residues of organophosphorus pesticides in the same reaction system.

Description

Technical field [0001] The invention belongs to the technical field of pesticide detection, and specifically relates to an immunoassay kit for detecting multi-residue organophosphorus pesticides and an application thereof. Background technique [0002] The multi-residue detection of small molecule chemical pollutants has always been the focus and difficulty of immunoassay. At present, there are mainly the following two models: (1) A model based on a single specific antibody. Separately prepare single specific antibodies against multiple pollutants, and distinguish different targets through different microwells on the microtiter plate or different arrays on the biochip, but the same reaction system cannot be detected simultaneously. (2) Model based on broad-spectrum specific antibodies. Only one antibody is needed to determine the total amount of multiple pollutants at the same time, but it cannot be quantified separately, and it may cross-react with non-target substances. Summ...
