[0020] In this study, 30 patients with histopathologically diagnosed bladder cancer (BUC) who underwent radical cystectomy (RC) from the Third Xiangya Hospital of Central South University and the Cancer Hospital Affiliated to Xiangya School of Medicine, Central South University from 2016 to 2018 were used. Buried specimens (Table 1). Six radical cystectomy (RC) BUC tissue specimens and corresponding adjacent non-neoplastic tissue specimens were immediately stored ...

Abstract

The research of the invention shows that lncRNA MAFG-AS1 is significantly overexpressed in primary BUC tissue, and the expression of MAFG-AS1 is up-regulated or down-regulated to enhance or inhibit the proliferation, metastasis and invasion of BUC cells in vitro and in vivo, respectively. Therefore, MAFG-AS1 can be used as a target site in the preparation of drugs for the treatment of bladder cancer. The mechanism study shows that MAFG-AS1 can directly bind to HuR and positively regulate HuR / PTBP1 axis to promote the proliferation, metastasis and invasion of BUC cells. Therefore, MAFG-AS1 canbe used in the preparation of targeted negative regulation reagent of HuR / PTBP1 axis expression.

Description

technical field [0001] The invention belongs to the technical field of biomedicine, and in particular relates to the application of MAFG-AS1 as a detection reagent of a target site in the preparation of a drug for treating bladder cancer. Background technique [0002] Bladder urothelial carcinoma (BUC) is the most common malignant tumor in the urinary system. In recent years, the morbidity and mortality have increased year by year. There are about 430,000 newly diagnosed cases worldwide each year, resulting in 165,000 patient deaths each year. Bladder cancer is divided into non-muscle invasive bladder cancer (NMIBC) and muscle invasive bladder cancer (MIBC), of which NMIBC accounts for 70% to 80% of bladder cancer cases, and its main features are rapid progression and easy metastasis, nearly 75% of High-grade NMIBC recurs, progresses, or dies within 5 years of diagnosis; MIBC usually requires radical cystectomy, combined with neoadjuvant chemotherapy or radiotherapy, but eve...
