[0165] Preparation of monoclonal antibodies

[0166] Antibodies of the present invention can be prepared by various techniques known to those skilled in the art. For example, an antigen of the invention, such as an EVD68 virus-like particle (VLP), can be administered to an animal to induce the production of monoclonal antibodies. For monoclonal antibodies, hybridoma technology can be utilized to prepare (see Kohler et al., Nature 256; 495, 1975; Kohler et al., Eur.J.Immunol.6:511, 1976; Kohler et al., Eur.J.Immunol. 6:292,1976; Hammerling et al., In Monoclonal Antibodies and T Cell Hybridomas, Elsevier, N.Y., 1981) or can be prepared by recombinant DNA methods (US Patent No. 4,816,567).

[0167] Representative myeloma cells are those that fuse efficiently, support stable high-level production of antibody by selected antibody-producing cells, and are sensitive to culture medium (HAT medium matrix), including myeloma cell lines, such as murine Myeloma cell lines, including tho...

Abstract

The invention discloses a neutralizing monoclonal antibody capable of resisting an enterovirus D68 in a broad-spectrum way. Particularly, the invention discloses two strains of monoclonal antibodies which aim at the enterovirus D68. The monoclonal antibodies have obvious binding and neutralizing activity for the enterovirus D68.

Description

technical field [0001] The invention belongs to the field of biomedicine, in particular, the invention relates to two kinds of neutralizing monoclonal antibodies against enterovirus D68. Background technique [0002] Enterovirus D68 (EVD68) is a small, non-enveloped virus belonging to group D of the genus Enterovirus within the family Picornaviridae. Similar to other picornaviruses, it has an icosahedral capsid of ~30nm, composed of 60 protomers, each of which consists of four subunit proteins, VP1, VP2, VP3 and VP4. Based on the evolutionary analysis of the VP1 nucleic acid sequence, the prevalent EVD68 strains can be divided into three main evolutionary lineages, A, B, and C. EVD68 has been increasingly associated with childhood acute respiratory disease and / or severe neurological disease, mainly acute flaccid myelitis (AFM). Over the past decade, EVD68 has spread widely throughout the world, continuously causing outbreaks and sporadic cases, thus posing a serious threat...
