Embodiment 1

[0044] This example shows a representative method of the invention for producing taeda pine (Pinus taeda) populations, cotyledon-type embryos, and germination of the embryos.

[0045] Large gametophytes containing zygotic embryos were removed from seeds 4-5 weeks after fertilization. The seed coat was removed, but the embryo was not further separated from the surrounding gametophyte other than the excision of the nucellar end. Store the cones at 4°C until use. Immediately before the removal of immature embryos, the seeds undergo a sterilization process: initial washing and detergent treatment, followed by 15% H 2 o 2 Sterilize for 10 minutes. Wash the explants thoroughly with sterile distilled water after each treatment.

[0046] Table 1 lists the composition of representative media used for taeda pine embryogenic tissues.

Abstract

The present invention provides methods for producing cotyledonary pine embryos. The methods of the invention each include the step of culturing embryogenic pine tissue in, or on, a medium comprising a disaccharide and glucose to yield cotyledonary pine embryos, wherein the disaccharide and glucose are each present in the medium at a concentration of less than 3 percent. The present invention also provides cotyledonary pine embryos prepared by a method of the invention.

Description

technical field [0001] The present invention relates to a method for producing plant embryos outside the plant and producing plants from the plant embryos arbitrarily. Background technique [0002] Demand for pine continues to rise for the manufacture of wood-based products. A proposed solution to this problem is to identify individual trees with desirable characteristics such as fast growth and obtain a large number of genetically identical clones of the superior tree through somatic cell cloning. These clones can be bred to obtain pine forests, or entire forests, with desired properties. [0003] A method for cloning pine trees is to process isolated living pine tree tissues in vitro under conditions that promote the formation of pine embryos, and then obtain whole plants from the treated tissues. Ex vivo pine tissue can be cultured in the presence of one or more auxins and / or cytokinins to promote the formation and proliferation of embryogenic tissue, followed by cultur...
