The method comprises the following steps: oxidizing tetravalent vanadium in back extraction water obtained in a total hydrometallurgical vanadium extraction process to form pentavalent vanadium; forming an oversaturated system of pentavalent vanadium, precipitating to obtain a polyvanadic acid precipitate, carrying out solid-liquid separation to separate a solution containing a high content of impurities, washing the polyvanadic acid precipitate, adding the washed precipitate to a sulfuric acid solution, carrying out heat insulation uniform stirring, and adding an ammonium bicarbonate solution to prepare a highly pure product, wherein polyvanadic acid gradually forms a polyammonium vanadate precipitate having a lower solubility due to the solubility of polyammonium vanadate is less than the solubility of polyvanadic acid; and calcining the polyammonium vanadate precipitate to generate vanadium pentoxide.
