Problems solved by technology

[0004] The purpose of this invention is to provide a new method for measuring biological activity such as biological pesticides. The method utilizes peach aphid as a test insect and agar layer as a source of nutrition and a moisture-retaining material for short-term cultivation of plant isolated parts in the biological activity measurement. Improves the accuracy of bioactivity testing due to problems with moisture retention methods such as filter paper or cotton wool

[0015] Comparison of the effects of filter paper moisture retention and agar moisture retention in the bioassay of biological samples

[0016] The biological sample (processing) used in Example 1 is a protein induced and expressed in bacteria by genetic engineering methods. In this example, the processing is the protein solution of the sample to be tested, and the control is other proteins that do not contain the processing sample. Repeat 4 times. In the group of moisturizing with filter paper, the mortality rate of the control group was 22.5%, and the mortality rate of the group treated with the bacterial solution reached 62.5%. There was a large difference between different treatments and controls; Reach 52.5%, and the mortality rate of matched group is 0% (table 1), handles or compares different repeated differences little, the result shows that the application of agar moisturizing of the present invention can effectively reduce the error that brings because humidity instabi...

Abstract

The invention relates to a novel method for measuring the biological activity of biological pesticides, which belongs to a novel method for measuring the biological activity. The novel method is characterized in that an agaragar layer is taken as a short-time cultivation nutrient source and a moisturizing material of a plant excised part in the measurement of biological activity; and meanwhile, green peach aphids are adopted as biological detecting materials to detect the biological activity of a detecting sample, and the insecticidal activity measurement is carried out according to the average reproduction quantity and the average death rate of the green peach aphides and the relevance of biological pesticides to be detected. The method solves the problems that plant leaves are dried easily and dehydrated generally in the insect bioassay by utilizing excised plant leaves to further influence the bioassay result and improves the accuracy of the experimental result. The method has simple and convenient operation, short experimental period and low cost. The invention can be applied to the biological activity measurement of biological pesticides and other pesticides of current varieties and the screening researches of new pesticides and is especially suitable for the bioassay of biological pesticides to insects with sucking mouth parts, such as aphides, and the like.

Description

technical field [0001] The invention relates to a kind of agar layer which is used as a nutrient source and moisture-retaining material for short-term cultivation of the isolated part of the plant in the determination of the biological activity of the biological pesticide. The biological activity of the aphid and the isolated part of the plant is used to measure the biological activity, which is especially suitable for the pricking and sucking of the aphid by the biological pesticide Determination of biological activity in insects with mouthparts. It belongs to the field of pesticide bioassay. Background technique [0002] There are two types of pesticide activity determination methods commonly used in the research and development of new pesticides: chemical assays and biological assays. The chemical determination method is based on some special reactions between chemicals and the test sample, such as: color reaction, etc., and evaluates the activity of the test sample thro...
