Problems solved by technology

[0002] For a long time, the traditional insect species identification technology is mostly based on the morphological characteristics of adults, which cannot realize the identification of non-adult states such as eggs, larvae and pupae, and it is difficult to distinguish similar species with subtle differences.

Embodiment 1

[0014] Example 1. Using specific primers to identify bookworms from stored booklices by PCR

[0015] (1) Extraction of Genomic DNA of Booklice

[0016] Genomic DNA of single-headed booklice from different geographic populations was extracted by CTAB method, including booklice from the Czech Republic and the United States, booklice from China, the Czech Republic, and the United States, and booklice from China and the Czech Republic Lice, Booklice microphthalmia collected from China, Czech Republic, USA, Booklice colorless from China, Czech Republic, USA, Booklice dunna collected from Czech Republic, USA, and Booklice hypocrisy collected from China. The species of the above booklice were confirmed by morphological characteristics.

[0017] Take a single head of the booklice to be checked and kill it with absolute ethanol, wash it with double distilled water and dry it (do not dry it out), put it into a 1.5mL sterile centrifuge tube, add 50μl extraction buffer (100mmol / L Tris-HC...

Abstract

The invention discloses a primer pair for identifying booklice in stored booklices and an application thereof. The nucleotide sequences of the primer pair for identifying booklice in stored booklices are respectively a sequence 1 and a sequence 2 in a sequence table. The invention also discloses a method for using the primer pair to identify the booklice in stored booklices. The method takes genome group DNA of the stored booklices to be measured as a template, uses the primer pair to carry out PCR amplification, and detects the PCR amplification product, and if the size of the PCR amplification product is 250 to 500bp, the stored booklice to be measured is the booklice. By the method for identifying booklice in stored booklices, the invention realizes fast and accurate identification of the booklice, distinguishes the booklice from other common stored booklices in the world, and has the characteristics of time saving, high efficiency and intuition and the like. The method can be widely used in inspection quarantine departments and grain storage departments.

Description

technical field [0001] The present invention relates to a pair of primers for identifying booklice from storage booklice and its application. Background technique [0002] For a long time, the traditional insect species identification technology is mostly based on the morphological characteristics of adults, which cannot realize the identification of non-adult states such as eggs, larvae and pupae, and it is difficult to distinguish similar species with subtle differences. Since the 1990s, with the development of modern molecular biology techniques, many scholars at home and abroad have explored molecular identification of various insects. When modern molecular biology is used to identify insects, the information of the gene sequence represents the genetic essence, which is not affected by the individual development status of the specimen and environmental conditions, and can provide sufficient quantitatively measurable genetic information. Therefore, it is an efficient and...
