The invention relates to an experimental method of micronucleus effect on vicia faba root tip cells by nano zinc oxide, sequentially comprising the following steps: a. soaking seed and accelerating germination; b. preparing contamination liquid; c. carrying out contamination; d. recovering for cultivation; e. fixing for dissociation; and f. carrying out microscopic examination and statistics.
