Problems solved by technology

[0005] The adverse effects of tobacco on human health have been widely reported all over the world. Finding stable and sensitive immune monitoring indicators is very important for monitoring and protecting the health of the population. Disease is of great significance, but there is still a lack of sensitive and stable immune evaluation indicators

Embodiment 1

[0029] Materials and Methods

[0030] 1. Materials

[0031] 1.1 Experimental animals, SD rats were purchased from Shanghai Xipuer-Bikai Experimental Animal Co., Ltd., license number SCXK (Shanghai): 2008-0016. No.1 low-tar cigarette (tar content 8mg / stick); No.2 ordinary cigarette (tar content 13mg / stick) was provided by the Technology Center of Zhejiang China Tobacco Industry Co., Ltd.

[0032] 1.2 Reagents: Rat IL-6ELISA kit and Rat TNF-αELISA kit were purchased from R&D Company, MTT Proliferation Detection Kit was purchased from SIGMA, Rat Ig ELISAkit was purchased from Lanji Biology, disposable cell counting plate was purchased from Handsome Company, and other reagents were Reagent grade reagents.

[0033] 1.3 Equipment: rat nasal exposure smoking system (refer to the CSES system of Battelle Company, Northwest Toxicology Research Center, USA), this system can be directly connected with JJS-II automatic smoking machine.

Embodiment 2

[0085] In addition to the experimental animals used: 80 healthy SD rats, each with a body weight of 150±10 grams, were randomly divided into 3 groups, group D, normal control group, inhaled air; group E, low-tar cigarette group, inhaled No. 3 Low-tar cigarettes (tar 8mg / stick); F group, common cigarette group, smoked No.4 common cigarette (tar 12mg / stick). Groups E and F smoked 20 cigarettes per day for 90 consecutive days with nasal smoking only, and stopped smoking for 42 days. No.3 low-tar cigarettes (tar content 8mg / stick); No.4 ordinary cigarettes (tar content 12mg / stick) were provided by the Technology Center of Zhejiang China Tobacco Industry Co., Ltd. Others are with embodiment 1.

[0086] The difference results between the smoking group and the normal control group are consistent with those in Example 1, which verifies the stability of the screening results of the screening method of the present invention.

[0087] Finally, five indicators were selected as sensitive...

Abstract

The invention discloses a screening method of immunologic function evaluation indexes of an anthropopathic nose-only rat. The screening method comprises the following steps: taking serum, splenic lymphocyte, splenic lymphocyte culture supernate, peritoneal macrophage and blood of an air breathing rat as a normal contrast group, and taking serum, splenic lymphocyte, splenic lymphocyte culture supernate, peritoneal macrophage and blood of a rat continuously smoking cigarettes through the nose as a smoking group; detecting the multiplication capacities of the lymphocyte, the swallowing conditions of the peritoneal macrophage, the contents of IL-6, TNF-alpha, Ig, IgG and IgE and the differential counting of leukocyte of each group; and finally, screening out five indexes of lymphopoiesis stimulation indexes, the contents of the IL-6, the IgG and the IgE and the differential counting of leukocyte as sensitive and stable evaluation indexes for reflecting the changes of the immunologic functions of the smoking rat. The method is scientific and comprehensive, and the screened evaluation indexes have good sensitive stability.

Description

technical field [0001] The invention relates to the field of immunological evaluation index screening, in particular to a method for screening immune function evaluation index of anthropomorphic smoking rats with only noses. Background technique [0002] Cigarette smoke is a complex mixture containing more than 6,000 chemical substances. Cigarette combustion is a very complex physical and chemical process. Due to the thermal decomposition and combustion of tobacco components, a large number of chemical substances will be generated, including CO, Free radicals, polycyclic aromatic hydrocarbons (PAHs), heterocyclic compounds, quinones, phenols, aldehydes, ketones, acids, esters, aliphatic compounds and other organic components, also contain trace amounts of inorganic elements such as Fe, Cu, Cr, Cd, Zn, etc. High concentrations of free radicals and chemical components that can react with other substances to generate active substances, such as CO, aldehydes, and polycyclic aro...
