Embodiment Construction

[0029] The present invention will be described in detail below according to the accompanying drawings, which is a preferred embodiment among various implementations of the present invention.

[0030] In a preferred embodiment, a method for preparing a manganese dioxide nanosheet and a super-sandwich-based fluorescent sensor comprises the steps of:

[0031] (1) The DNA sequence to be purchased (HP 1 , HP 2 , helperDNA) was dissolved in PBS (pH7.4) buffer solution, and stored at low temperature for future use, HP 1 , HP 2 After heating at 80-90°C for 10 minutes, cool naturally to room temperature and incubate for a period of time;

[0032] (2) The substances used such as manganese dioxide nanosheets, coralyne, heparin, and SG are configured into corresponding concentrations;

[0033] (3) HP 1 , HP 2 , helperDNA, coralyne, SG, and manganese dioxide nanosheets were added to the solution and incubated at 45°C for 10 minutes;

[0034] (4) The concentration of heparin is diffe...

Abstract

The invention relates to a fluorescent sensor, a preparation method thereof and an application of the fluorescent sensor, in particular to a biological fluorescent sensor based on manganese dioxide nano-sheets and super-sandwiching. By the characteristics that fluorescent light can be quenched by the manganese dioxide nano-sheets, single-stranded DNA (deoxyribonucleic acid) can be adsorbed on the manganese dioxide nano-sheets, and ds DNA and other DNA with a secondary structure can be far away from the manganese dioxide nano-sheets, the fluorescent sensor can be used for detecting heparin. The preparation method of the fluorescent sensor comprises the following steps of (1) dissolving a DNA sequence (HP1, HP2 and helper DNA) in buffered solution and storing the buffered solution for future application; (2) preparing used substances until the concentration is suitable; and (3) culturing the HP1, the HP2, the helper DNA and the used substances in the solution. In the preparation method of the biological fluorescent sensor, unmarked DNA is used. The preparation method is easy to operate and quite low in cost, chemical marks and modification are avoided, and a result shows that the detection result of the sensor on the heparin is satisfactory. The fluorescent sensor has the advantages of easiness in operation, high sensitivity and low detection limit.

Description

technical field [0001] The invention belongs to the technical field of biosensors based on two-dimensional nanomaterial manganese dioxide nanosheets, and specifically relates to the construction of a fluorescence sensor based on two-dimensional nanomaterial quenching of fluorescence and super-sandwich induced by DNA to trigger luminescence signals for the detection of heparin detection. Background technique [0002] The 21st century is the century of life science, and the study of DNA is an extremely important aspect in life science research, and the research of DNA biosensor has become a hot spot. At present, various methods are used to detect biomolecules. The emergence of two-dimensional nanomaterials has injected strong vitality into DNA-based sensors. The combination of two-dimensional nanomaterials and DNA-based sensors is a product of the times. Two-dimensional manganese dioxide nanosheets have been studied due to their good water solubility, excellent biocompatibili...
