Abstract

The invention relates to a method for extracting and preparing a blood-sugar-reducing substance from the seeds of Fraxinus rhynchophylla Hance. The method comprises the following concrete steps: crushing the seeds of Fraxinus rhynchophylla Hance and subjecting the crushed seeds to percolation and extraction with 75% ethanol twice so as to obtain a 75% ethanol extraction solution, wherein the seeds are soaked for three days in each percolation and extraction; carrying out micro-membrane filtration on the 75% ethanol extraction solution and then carrying out HPLC detection; and concentrating 75% ethanol extraction liquid so as to obtain extract, dispersing the extract with water, carrying out extraction with petroleum ether, concentrating a petroleum ether layer solution so as to obtain another extract, laying the extract aside, extracting a water layer with ethyl acetate, concentrating an ethyl acetate layer so as to obtain another extract, carrying out petroleum ether-acetone gradient elution, wherein TLC detection and analysis are cooperatively carried out in the process of elution, flushing a column with ethanol, combining eluate of a same part and carrying out respective drying by distillation so as to obtain separated compounds, wherein the separated compounds include secoiridoid glucoside compounds. The method provided by the invention has high extraction rates of components in the seeds of Fraxinus rhynchophylla Hance and can effectively extract the secoiridoid glucoside compounds capable of reducing blood sugar.
