Embodiment 1

[0038] This example provides a method for simultaneous detection of total mercury, inorganic mercury and organic mercury in tobacco. The schematic diagram of the detection route is shown in the attached figure 1 shown, including the following steps:

[0039] S1. Sample pretreatment: Grind the tobacco sample and pass it through a 40-mesh sieve to obtain the sample to be tested. Accurately weigh 0.2g of the sample to be tested into a 15mL centrifuge tube, add 10.00mL of 5mol / L hydrochloric acid as the extraction solution, shake well and soak overnight (8-12 hours). Then 40kHz, 100W ultrasonic extraction for 30min, 4000r / min centrifugation for 30min, the obtained supernatant was passed through a 0.45μm water filter membrane to obtain the filtered supernatant for later use;

[0040] S2. Detect the mass fraction of inorganic mercury:

[0041] Take 3.00 mL of the filtered supernatant from S1 in a 10 mL colorimetric tube, add 0.5 mL of n-propanol to eliminate air bubbles, and dilu...

Embodiment 2

[0054] This example provides a method for simultaneous detection of total mercury, inorganic mercury and organic mercury in tobacco. The schematic diagram of the detection route is shown in the attached figure 1 shown, including the following steps:

[0055] S1. Sample pretreatment: Grind the tobacco sample and pass it through a 40-mesh sieve to obtain the sample to be tested. Accurately weigh 0.2g of the sample to be tested in a 15mL centrifuge tube, add 15.00mL of 4mol / L hydrochloric acid as the extraction solution, shake well and soak overnight. Then, it was extracted by 40kHz, 100W ultrasonic wave for 40min, centrifuged at 4000r / min for 30min, and the obtained supernatant was passed through a 0.45 μm water filter membrane to obtain the filtered supernatant for later use;

[0056] S2. Detect the mass fraction of inorganic mercury:

[0057] Take 1.5 mL of the filtered supernatant from S1 in a 10 mL colorimetric tube, add 0.5 mL of n-propanol to eliminate air bubbles, and ...

Embodiment 3 application example 1

[0064] Determination of organic mercury in raw tobacco leaf samples: the method of the present invention was applied to the determination of total mercury, inorganic mercury and organic mercury in 5 kinds of raw tobacco leaf samples used by the applicant unit, and the results showed that no inorganic mercury or organic mercury was detected.

Abstract

The invention discloses a method for detecting total mercury, inorganic mercury and organic mercury in tobacco simultaneously and an application. One-time sample pretreatment is combined with cold vapor generation-atomic fluorescence spectrometry in the same detection condition for detecting the mass fraction of inorganic mercury and detecting the mass fraction of total mercury respectively, and the mass fraction of organic mercury is obtained by calculation, so that the content of total mercury, inorganic mercury and organic mercury in tobacco can be detected simultaneously by one-time sample pretreatment. The invention provides a simultaneous analysis detection scheme of mercury at different states in tobacco, the accurate detection result of total mercury, inorganic mercury and organic mercury can be obtained without form fractionation, and based on the method provided by the invention, the form distribution of mercury in tobacco can be reflected subjectively. The method is simple and convenient to operate, is high result accuracy and flexibility, and is wide in linear wide; the used instrument is simple and is easy to popularize.

Description

technical field [0001] The invention relates to the technical field of analysis of inorganic elements in tobacco, and more particularly relates to a method and application for simultaneously detecting total mercury, inorganic mercury and organic mercury in tobacco. Background technique [0002] Mercury has three valence states of zero valence, positive one valence and positive two valence, and can form various forms of compounds with various organic and inorganic ligands. The order of toxicity of different forms of mercury from strong to weak is organic mercury, inorganic mercury and elemental mercury. Plants mainly contain inorganic mercury (II) and organic mercury, of which organic mercury is represented by methylmercury (CH 3 Hg + ) and ethylmercury (C 2 h 5 Hg + ) mainly. [0003] At present, there have been many technical reports on the total determination method of mercury in tobacco, but there is no report on the speciation analysis method of mercury in tobacco....
