Embodiment 1

[0021] Preparation of standard product: After opening the 5g standard product of CuCl, divide it into 10 1mL plastic tubes, carefully fill with argon gas, drive the air out of the tube, seal the tube mouth with sealant, and store it in a desiccator with silica gel middle.

[0022] A method for measuring monovalent copper content by liquid-liquid extraction-GFAAS method, comprising the steps:

[0023] S1. Preparation of the standard working curve: When preparing the working curve, take out one of the tubes, weigh 0.0156g of the standard CuCl (99.999%, Aladdin) and place it in a 10mL volumetric flask, and dilute it to the mark with saturated KCl solution to obtain 1000ppm sub-Cu standard solution, take 100μL from it and put it in a 5mL plastic tube, add 900μL pH 9Glycine (glycine)-NaOH buffer solution, add 1000μL 0.06% 2,2'-biquinoline n-amyl alcohol solution, vortex for 1min , standing and layering, the upper layer is 100ppm cuprous standard solution, then take 100μL from the ...

Abstract

The invention belongs to the technical field of trace element detection and particularly discloses a method of measuring the content of monovalent copper through liquid liquid extraction-GFAAS. The method includes S1) a step of making a standard working curve, and S2) a step of detecting the monovalent copper, namely a step of fully mixing a sample and trichloroacetic acid the concentration of which is 20% and the volume of which is the same as the volume of the sample, diluting with a glycine-NaOH buffer solution having a pH value of 9 according to different ratios according to the copper content of the sample, adding 1 mL of an n-pentanol solution of 2,2'-biquinoline with a concentration of 0.06% (w / w) into the solution mixture, fully mixing, allowing the mixture to stand and layer, obtaining an upper organic layer, and measuring the content of the monovalent copper by subjecting the upper organic layer to graphite furnace atomic absorption spectrometry. The method can be used for detecting the content of trace monovalent copper in biological samples, and is of great importance in research on organic body copper metabolism imbalance.

Description

technical field [0001] The invention belongs to the technical field of trace element detection, and in particular relates to a liquid-liquid extraction-GFAAS method for determining the content of monovalent copper. Background technique [0002] Copper ions are indispensable trace elements for organisms. In organisms, copper mainly exists in the state of complexing with enzymes, protein molecules and some small biological molecules such as amino acids. Copper ions participate in some important physiological processes of organisms, such as cellular respiration, transmission of neurotransmitters, anti-oxidative stress and uptake of iron ions are all dependent on copper ions. Many diseases in the human body are related to the imbalance of copper ions in the body, such as hepatolenticular degeneration, mad cow disease, Alzheimer's disease and cancer. Neurodegenerative diseases are associated with loss of ceruloplasmin bioactivity or Cu(I) toxicity. The role of copper ions in l...
